ABSTRACT Background and Aim Metabolic dysfunction associated steatotic liver disease (MASLD) is the predominant cause of chronic disease, with dysregulation bile acid (BA) metabolism intestinal microbiota being intricately MASLD progression. In this study, we investigated role ileal FXR in progression BA portal blood. Methods Sprague–Dawley rats were fed a typical western diet for 20 weeks, followed by local perfusion AAV2‐sh Nr1h4 to downregulate expression ileum tissue. To investigate effect on reabsorption gut microbiota, blood cecal fecal samples collected from injected AAV2‐Ctrl or metabolomics targeting BAs 16S rRNA sequencing analysis. Results Our results showed that hepatic steatosis inflammation alleviated, whereas secondary unconjugated into was enhanced when knocked down. Furthermore, knockdown resulted significant alteration composition characterized an increasing abundance microbes involved production, including Escherichia , Adlercreutzia Eubacterium Clostridium . Conclusion These findings suggest downregulation ameliorates modulating mediated indicating might be potential therapeutic target treatment MASLD.
