Background Tenebrionidae (Insecta: Coleoptera) are a conspicuous component of desert fauna worldwide. In these ecosystems, they significantly responsible for nutrient cycling and show remarkable morphological physiological adaptations. Nevertheless, colonizing individual deserts have repeatedly emerged from different lineages. The goal our study was to gain insights into the phylogenetic relationships tenebrionid genera Atacama Desert how taxa related globally distributed Tenebrionidae. Methods We used newly generated transcriptome data (47 tribes, 7 11 subfamilies) that allowed comprehensive phylogenomic analysis this hyperarid fills gap in knowledge highly diversified examined two independent sets known be suitable reconstructions. One is based on 35 neuropeptide precursors, other 1,742 orthologous genes shared among Coleoptera. Results majority placed three groups, which belong typical South American lineages within Pimeliinae. While support monophyly Physogasterini, Nycteliini Scotobiini, does not hold Edrotini, Epitragini, Evaniosomini, Praociini, Stenosini, Thinobatini, Trilobocarini. A suggested very close relationship Psammetichus with Mediterranean Leptoderis also could confirmed. provide hints regarding Caenocrypticini, occur both America southern Africa. Apart focus Desert, we found striking synapomorphy grouping Alleculinae, Blaptinae, Diaperinae, Stenochinae, several Tenebrioninae, but Tenebrio Tribolium . This character, an insertion myosuppressin gene, defines higher-level monophyletic group Conclusion Transcriptome allow represents one seven major endemic tribal areas world Most America; supported tribes molecular data, suggesting detailed systematic revision groups necessary.

Abstract The short neuropeptide F (sNPF) family of peptides is a multifunctional group neurohormones involved in the regulation various physiological processes insects. They have been found broad spectrum species, but number isoforms precursor molecule varies from one to four. receptor for sNPF (sNPFR), which belongs G protein-coupled family, has characterized insect orders and was shown be an ortholog mammalian prolactin-releasing peptide (PrPR). signaling pathway interacts with other such as insulin-like peptides, SIFamide, pigment-dispersing factors (PDFs) regulate processes. main function seems feeding, observed effects are species-specific. also connected foraging behavior olfactory system. influence on feeding thus energy metabolism may indirectly affect vital processes, reproduction development. In addition, these locomotor activity circadian rhythm This review summarizes current state knowledge about system

Adipokinetic hormone (AKH) is a neuropeptide produced in the insect corpora cardiaca that plays an essential role mobilising carbohydrates and lipids from fat body to haemolymph. AKH acts by binding rhodopsin-like G protein-coupled receptor (GPCR), adipokinetic (AKHR). In this study, we tackle ligand gene evolution as well evolutionary origins of paralogues order Blattodea (termites cockroaches). Phylogenetic analyses precursor sequences point ancient duplication event common ancestor Blaberoidea, yielding new group putative decapeptides. total, 16 different peptides 90 species were obtained. Two octapeptides seven putatively novel decapeptides are predicted for first time. 18 species, spanning solitary cockroaches subsocial wood roaches lower higher termites, subsequently acquired using classical molecular methods silico approaches employing transcriptomic data. Aligned AKHR open reading frames revealed 7 highly conserved transmembrane regions, typical arrangement GPCRs. based on support accepted relationships among termite, (Cryptocercus spp.) cockroach lineages large extent, while post-translational modification sites do not greatly differ between social termites. Our study provides important information only functional research but also further interested their development potential candidates biorational pest control agents against invasive termites cockroaches.

Abstract Neuropeptides are crucial in regulation of a rich variety developmental, physiological, and behavioral functions throughout the life cycle insects. Using an integrated approach multiomics, we identified neuropeptide precursors greater wax moth Galleria mellonella , which is harmful pest honeybee hives with worldwide distribution. Here, total 63 67 were predicted annotated G. genome transcriptome, 40 confirmed peptidome. Interestingly, 12 precursor genes present but absent honeybees, may be potential novel pesticide target sites. Honeybee contaminated heavy metals such as lead, enabling its bioaccumulation bodies through food chain, performed transcriptome sequencing to analyze effects Pb stress on mRNA expression level precursors. After treatment by Pb, F1 was found significantly downregulated, implying that this might associated responding metal . This study comprehensively discussed insect neuropeptides, example The results valuable for future elucidation how neuropeptides regulate physiological contribute our understanding insect's environmental plasticity identify new biomarkers assess toxicity

The central nervous system (CNS) plays a critical role in signal integration animals and allows the orchestration of life processes to maintain homeostasis. Current research clearly shows that inflammatory can also be modulated by CNS via neuroendocrine system. One neuropeptide families participate vertebrates this process is orexins (OXs). Interestingly, our previous results suggested similar dependency may exist between neuropeptides immune activity insects. Due structural homology orexin allatotropin receptors functional similarity these two families, main aim was perform complex analysis relationships (AT) insect response. Our revealed similarities vertebrate OXs ATs. Similar effects were observed profile expression level gene encoding AT precursor Tenebrio molitor general action Tenmo-AT on selected parameters tested beetles. Moreover, for first time insects, we confirmed cytokines modulation determining effect Spätzle-like protein injection genes receptor. Tachykinin-related peptides (TRPs) are one of the most prominent families neuropeptides in animal kingdom. Insect TRPs display strong structural and functional homology to vertebrate tachykinins (TKs). To study homologies between these two neuropeptide families, influence human substance P (SP, essential TKs) on immune system mealworm beetle, Tenebrio molitor L., was analysed. Human SP influences phagocytic abilities T. haemocytes. Peptide injection leads an increase number haemocytes participating phagocytosis latex beads. In contrast, incubation from non-injected beetles a solution physiological saline causes decrease activity. Treatment with also led increased adhesion haemocytes, but no changes arrangement F-actin cytoskeleton were observed. Interestingly, 6 h after injection, DNA integrity reported. The opposite effects observed 24 injection. caused upregulation humoral responses, such as phenoloxidase (PO) activity haemolymph, downregulation immune-related genes encoding coleoptericin A, tenecin 3 Toll receptor. However, attacin 2 cecropin upregulated. Despite differences, antimicrobial haemolymph significantly lower injected than control beetles. Moreover, analysis direct lysozyme performed. Our results suggest that at concentration 10−6 M can directly inhibit effect reported application 10−4 M. presented TK signalling vertebrates insects. Primarily, this visible context response general haemolymph. some related haemocyte function may indicate importance TRP sequences for evoking immunological effects.
