“True” cobras (genus Naja) are among the venomous snakes most frequently involved in snakebite accidents Africa and Asia. The Cape cobra (Naja nivea) is one of African highest medical importance, but much remains to be learned about its venom. Here, we used a shotgun proteomics approach better understand qualitative composition N. nivea venom tested cytotoxicity protease activity as well effect on intracellular Ca2+ release NO synthesis. We identified 156 components representing 17 protein families, with dominant ones being three-finger toxins, mostly short-chain type. Two-thirds toxin entries were assigned cytotoxins, while remainder categorized neurotoxins, including short-chain, long-chain, ancestral toxins. also snake metalloproteinases members CRISP, l-amino acid oxidase, other families. Protease synthesis low. Phospholipase A2 was surprisingly high, despite this family marginally recovered analyzed Cytotoxicity relevant only at higher concentrations, macrophage neuroblastoma cell lines showing lowest viability.
