As the most abundant protein in blood, albumin binds and transports hydrophilic, hydrophobic, lipophilic molecules required for growth development of cells tissues. Due to surface location charged groups inner hydrophobic site, it proteins, peptides, amino acids, fatty nutrients, other biomolecules. It is a multi-domain involved various functions including contribution oncotic pressure. Because its unique sequence structure, can be efficiently utilized develop drug delivery systems against diseases, cancer. protein, rich both negative positive charges on surface, attracts great attention discipline. Inner sites were proven an efficient delivering drugs. Albumin binding domains, widely distributed among receptors matricellular molecules, ease penetration into cells. A high ratio cysteine provides stability molecule causes thiol-disulfide interactions, which play essential role release. In this work, we highlighted several points nature as drug-carrying regarding biochemical physical properties: self-assembling, formation covalent bonds, situ corona, etc. Research publications searched NCBI database by keywords related sections/subsections review topic. Tens research works have been dedicated enhancing therapeutic efficacy anticancer drugs using last two decades contributed clarifying mechanistic action albumin. We discussed mechanisms first section. another section, reviewed enhanced antitumor/anticancer efficacies those fabricated DDSs terms contribution. Human serum albumin (HSA) is the most abundant plasma protein of circulatory system. It a multidomain, multifunctional that, combining diverse affinities and wide specificity, binds, stores, transports variety biological compounds, pharmacores, fatty acids. HSA finding increasing uses in drug-delivery due to its ability carry functionalized ligands prodrugs. All this raises question competition for binding sites occupancy case multiple ligands, which turn influences structure/dynamic/function relationship also has an impact on biomedical applications. In work, effects interactive palmitic acid (PA), warfarin (War) ibuprofen (Ibu) thermal stability were studied using DSC, ATR-FTIR, EPR. PA high-affinity physiological ligand, while two drugs are widely used their anticoagulant anti-inflammatory efficacy, exogenous compounds that accommodate deputed drug site DS1 DS2, respectively overlapping with some sites. The results indicate acquires highest when it fully saturated PA. ligand does not hamper War or Ibu native state protein. addition, three bind simultaneously, suggesting synergic cooperative influence allosteric effects. increased subsequent binary moderates aggregation propensity restricts dynamics. biophysics findings provide interesting features about stability, aggregation, dynamics interaction relevant drug-delivery.
