Highly efficient recognition of cancer cells by immune is important for successful therapeutic-cell-based immunotherapy. Herein, we present a facile NIR-II nanoadaptor [hyaluronic acid (HA)/dibenzocyclooctyne (DBCO)-Au:Ag2Te quantum dots (QDs)] enhancing the tumor and binding ability natural killer (NK) via bio-orthogonal click reaction in vivo. The Nanoadaptor possesses superior tumor-targeting capacity, facilitating accumulation chemical receptor DBCO at sites. Abstract Pancreatic cancer is a malignancy tumor with luxuriant extracellular matrix (ECM) and highly immunosuppressive microenvironment its therapy remains an enormous challenge. A two‐step targeting‐tunable strategy proposed for orthotopic pancreatic via designing semiconducting nanoswitches (C/SPN T /αP) to amplify the mitochondrion damage programmed death ligand 1 (PD‐L1) blockade. mitochondrion‐targeting small nanoparticle (SPN ) consisting of mitochondrial targeting moiety triphenylphosphine (TPP) polymer embedded PD‐L1 antibody (αPD‐L1) into sono‐responsive collagen binding peptide (CBP)‐conjugated nanoliposomes form C/SPN /αP. In first step, /αP achieve ECM observably increase their enrichment sites because CBP can effectively bind in ECM. Upon ultrasound (US) irradiation, mediate structural failure sonodynamic effect on‐demand releases SPN αPD‐L1. second target mitochondria induce under US irradiation triggering amplified cell apoptosis immunogenic (ICD). Moreover, released αPD‐L1 blocks pathway further boost immunological effect. Such novel therapeutic regimen almost completely eradicate Panc02 KPC tumors mouse models.
