Abstract Cardiac glycosides (CGs) are a class of bioactive organic compounds well-known for their application in treating heart disease despite narrow therapeutic window. Considerable evidence has demonstrated the potential to repurpose CGs cancer treatment. Chemical modification these been utilized attempts increase anti-cancer properties; however, this met limited success as mechanism action is still speculative. Recent studies have identified DNA damage response (DDR) pathway target CGs. DDR serves coordinate numerous cellular pathways initiate cell cycle arrest, promote repair, regulate replication fork firing and protection, or induce apoptosis avoid survival cells with carrying mutations. Understanding modus operandi cardiac will provide critical information better address improvements potency, reduced toxicity, overcome drug resistance. This review summarizes recent scientific findings molecular mechanisms affecting signaling therapeutics from 2010 2022. We focus on structural functional differences toward identifying features targeting agents.

This work describes the studies on direct C3-glycosylation of C19-hydroxylated cardiotonic steroids strophanthidol, anhydro-ouabagenin, and ouabagenin using a strategy based in situ protection C5 C19 hydroxyl groups with boronic acids. While this resulted successful one-pot C3-selective glycosylation strophanthidol anhydro-ouabegenin, it failed to provide ouabain from ouabagenin. The neuroprotective activity synthetic natural glycosides against LPS-induced neuroinflammation was explored neonatal mouse primary glia cells. Co-administration C3-glycosides at 200 nM concentrations significant reduction neuroinflammatory markers IL-6, IL-1, TNFα, IKBKE, anhydro-ouabagenin-3-(α)-l-rhamnoside (anhydro-ouabain) showing most effect. At same time, unglycosylated anhydro-ouabagenin enhanced rather than suppressed neuroinflammation.

A combination of DFT calculations and experiments is used to describe how the selection a promoter can control stereochemical outcome glycosylation reactions with deoxy sugar saccharosamine. Depending on promoter, either α- or β-linked reactive intermediates are formed. Carbohydrates are natural, renewable, chemical compounds that play crucial roles in biological systems. Thus, efficient and stereoselective glycosylation is an urgent task for the preparation of pure structurally well-defined carbohydrates. Photoredox catalysis has emerged as a powerful tool carbohydrate chemistry, providing alternative addressing some challenges glycochemistry. Over last few decades, Ir- Ru-based organometallic photocatalysts have attracted significant interest because their high stability, high-energy triplet state, strong visible-light absorption, long luminescence lifetime, amenability to ligand modification. This review highlights recent progress photocatalyst-promoted synthesis modification carbohydrates under photoirradiation, well related benefits drawbacks.
