Rheumatoid arthritis (RA) is one of the leading causes disability due to autoimmune destruction synovial joints. Great efforts have been put into developing multifunctional drug delivery systems for delaying progression RA. Herein, a novel locally injectable, hierarchically structured system with hyaluronic acid (HA) microparticle and an MTX‐conjugated nanoparticle promoted RA treatment proposed. In this system, MTX chemically grafted onto peptide dendrimer‐based (G‐MTX). These nanoparticles are encapsulated inside HA microparticles (H‐G‐MTX) using microfluidics realize system. The G‐MTX can be released from degradable in acidic environment inflamed As shows enhanced intracellular conjugated MTX, H‐G‐MTX reverse dominant macrophage phenotype M1 M2. Through collagen‐induced (CIA) rat model, great therapeutic outcomes demonstrated, including inflammation reduction, cartilage protection, bone mineral density promotion. results indicate that proposed hierarchical has potential clinical value other diseases.

In this study, manganese-doped albumin-gelatin composite nanogels (MAGN) were prepared and used to load berberine (Ber) for the treatment of gouty arthritis (GA). The nanodrug delivery system (Ber-MAGN) can target inflammatory joints due intrinsic high affinity albumin SPARC, which is overexpressed at site GA. Characterization pharmaceutical properties in vitro showed that Ber-MAGN had good dispersion, particle size was 121 ± 10.7 nm. sustained release effect significantly improved bioavailability berberine. vivo experimental results has better therapeutic effects relieving oxidative stress suppressing inflammation. Therefore, Ber-MAGN, as a potential preparation GA, provides new reference clinical plan
