Abstract Over the past decade, significant advancements have been achieved in field of biomedicine, particularly treatment critical illness. However, there remain numerous challenges and barriers persist drug delivery, medical imaging, diagnosis, treatment. Poly(amidoamine) (PAMAM) dendrimers emerged as promising candidates for development functional biomedical materials due to their advantageous properties, including ease preparation, controllable size, abundant cavities, water solubility, modifiability, biocompatibility. This article presents a comprehensive review synthesis, application, potential toxicity PAMAM materials, exploring role, significance, applications detail. The aim is provide valuable reference future inspire further discoveries polyamide materials.

Abstract Background Temporomandibular joint osteoarthritis (TMJ-OA) is a disease characterized by cartilage degradation and synovial inflammation, with limited effective treatment currently. Synovial macrophage polarization pivotal in TMJ-OA progression, making it promising therapeutic aspect. This study investigated the effects of S-propargyl-cysteine (SPRC), an endogenous H2S donor, on its potential alleviating TMJ-OA. Methods A MIA-induced rat model LPS-stimulated RAW264.7 macrophages were employed to evaluate SPRC vivo vitro. TMJ bone analyzed via micro-CT histological methods, while markers expression assessed RT-qPCR, western blot, immunofluorescence. RNA sequencing was performed macrophages, JAK2/STAT3 signaling pathway validated using JAK2-specific inhibitor AG490. The direct primary condylar chondrocytes examined evaluating ECM synthesis degradation. Co-culture experiments further macrophage-chondrocyte interactions. Results significantly alleviated damage model, as demonstrated improved volume structure. reduced pro-inflammatory M1 infiltration enhanced anti-inflammatory M2 polarization. effectively inhibited JAK2/STAT3, leading reduction inflammatory markers, including TNF-α, IL-6, iNOS. revealed that SPRC-treated macrophage-conditioned medium chondrocyte metabolic activity restored integrity. Conclusions SPRC-modulated alleviates JAK/STAT downregulation, thereby reducing inflammation These findings position candidate for provide insights into novel strategies targeting synovium-cartilage crosstalk. Cell-free DNA (cfDNA) holds significant promise for diagnostic and therapeutic advancements in medicine. This review delineates the utility of cfDNA diagnostics its potential through clearance mechanisms an array diseases. Damage-associated molecular patterns (DAMPs) are endogenous molecules released by host cells during stress, or injury. As a trigger inflammatory responses via damage-associated (DAMPs), cfDNA's removal nanotechnological approaches can attenuate inflammation promote tissue repair. While application is particularly auspicious cancer, sepsis, conditions, it confronted with challenges including toxicity, specificity, rigors clinical trial validation. Collectively, this novel targets to inform development innovative treatment strategies.
