Arsenic (As) is a metalloid naturally present in the environment, food, water, soil, and air; however, its chronic exposure, even with low doses, represents public health concern. For long time, As was used as pigment, pesticide, wood preservative, for medical applications; industrial use has recently decreased or been discontinued due to toxicity. Due versatile applications distribution, there wide spectrum of human exposure sources, mainly contaminated drinking water. The fact that water implies this metalloid; it become worldwide problem, since over 200 million people live where levels exceed safe ranges. Many problems have associated including cancer, cardiovascular diseases, gastrointestinal disturbances, brain dysfunctions. Because can cross blood–brain barrier (BBB), target organ exert long-term toxic effects. mechanisms neurotoxicity described: oxidative stress, inflammation, DNA damage, mitochondrial dysfunction; all them converge, thus leading impaired cellular functions, cell death, consequence, detrimental Here, we provide current overview toxicity integrated global involved cognitive behavioral impairment induced by show experimental strategies against neurotoxicity.

Functional polysaccharides are characterized by functional and structural biomolecules, which play a dual role, both as nutrient molecules providing energy signalling involved in intercellular communications. There increasing evidences showing that possess beneficial effects for treating chronic diseases, such cancer, cardiovascular disease, etc, but its mechanism of action remains imprecision. Additionally, due to the limitations precision detection methods complexity their structures, metabolic kinetics behaviours or bioavailability polysaccharides, interaction between it entering systemic circulation target cells have been conundrum faced researcher. This review summarized latest advances efficacy diseases structure-activity relationship, absorption behaviours, specific cellular receptors so understand mechanisms bioactivities. is expected provide new insights paves way application.

Chemical-induced neurotoxicity is increasingly recognized to accelerate the development of neurodegenerative disorders (NDs), which pose an increasing health burden society. Attempts are being made develop drugs that can cross blood–brain barrier and have minimal or no side effects. Nobiletin (NOB), a polymethoxylated flavonoid with anti-oxidative anti-inflammatory effects, has been demonstrated be promising compound treat variety NDs. Here, we investigated potential role NOB in sodium arsenate (NA)-induced deregulated miRNAs target proteins human neural progenitor cells (hNPCs). The proteomics microRNA (miRNA) profiling was done for different groups, namely, unexposed control, NA-exposed, NA + NOB, groups. Following correlation analysis between proteins, RT-PCR used validate selected genes. proteomic showed significantly were associated neurodegeneration pathways, response oxidative stress, RNA processing, DNA repair, apoptotic process following exposure NA. OpenArray confirmed altered regulate P53 signaling, Wnt cell death, cycle pathways. validation studies concur data as marker genes autophagy apoptosis (HO-1, SQSTM1, LC-3, Cas3, Apaf1, HSP70, SNCA1) exposure. It observed treatment restored their basal levels. Hence, it may considered one its neuroprotective mechanisms. Together, findings demonstrate applicability neuroprotectant against chemical-induced neurotoxicity.

Arsenic is a ubiquitous environmental toxin that can affect normal physiological processes. Although the health impacts of arsenic have been investigated, its influence on hepatic metabolism in obese pregnant women and underlying mechanisms remain unclear. Multi-omics analysis, including metabolomics proteomics, improve understanding arsenic-induced hepatotoxicity women. This study aimed to investigate adverse effects gestational exposure high-fat-diet-induced mice. Following during pregnancy, liver tissue was evaluated comprehensively using proteomics techniques combined with pathological biochemical analyses. not only significantly increased lipid accumulation livers mice but also elevated inflammatory factors oxidative stress markers. Specifically, histopathological examination revealed more steatosis, cell infiltration, hepatocyte ballooning arsenic-exposed than those controls. These changes indicate exacerbates induces damage context obesity. Metabolomic analysis provided further insight into metabolic-level disruption caused by exposure. Significant were observed pathways, particularly arachidonic acid pathway. As metabolites play important roles inflammation stress, this pathway may be critical hepatotoxicity. Additionally, proteomic showed differences expression levels several key proteins involved synthesis, response. Notably, oxidative-stress-related proteins, glutathione peroxidase 4 (GPX4), upregulated, suggesting an burden. In summary, there are complex interaction among exposure, response, related metabolism. The integration aided clarifying molecular alterations induced arsenic. results show affects through multiple metabolic pathways protein regulatory mechanisms. addition providing new insights relationship between obesity as well diseases, act reference for risk assessment formulation public policies. enhanced will contribute development strategies mitigating risks associated toxins, vulnerable groups such

Abstract Background Recent research showed that the NLRP3 inflammasome was activated in central nervous system of mice administered chronic ethanol (EtOH). Dictyophora polysaccharides (DIPs) are essential components valuable edible fungus , which has antioxidant properties can delay aging process body. This study aimed to investigate roles EtOH‐induced cerebellar Purkinje cell (PC) degeneration and behavioral changes. Methods C57BL/6J normal knockout were exposed EtOH for 14 days. polysaccharide (DIP) inhibitor mice. The pathology NLRP3‐ASC‐caspase‐1 signaling pathway proteins analyzed tissues performance assessed Results In mouse model, we observed increases proteins, including NLRP3, ASC, caspase‐1, mature IL‐1β pro IL‐1β, loss PCs, motor coordination disorders. We found DIPs could suppress pathway, alleviate deficits pathological changes Next, used MCC950, a inhibitor, an strategy further verify effects MCC950 or alleviated latency decreases fall time, stride width length. also attenuated PC number suppressed inflammation induced by EtOH. Taken together, pharmacologically genetically inhibiting deficits. Conclusion These findings indicated might diminish through provides potential therapeutic target prevention treatment alcoholism pathology.
