Supramolecular Self‐Assembly of Monosubstituted Pillar[5]Arenes Under the Control of the Nature of the Amide Moiety

Abstract
In
this
study,
we
have
shown
for
the
first
time
that
introduction
of
a
single
alkylamide
substituent
into
macrocyclic
platform
pillar[5]arene
opens
up
possibility
to
control
supramolecular
properties
platform.
The
ability
synthesized
pillar[5]arenes
form
pseudorotaxane
associates
both
in
solutions
and
crystalline
state
was
studied
by
complex
physical
methods.
Linear
secondary
amide
fragments
such
as
N
,
‐diethylethane‐1,2‐diamide,
‐dimethylpropane‐1,3‐diamide,
or
‐aminoethylmorpholide
structure
promote
formation
self‐inclusion
complexes
solution
states.
Whereas
tertiary
amides
pyrrolidide
morpholide
moieties
favor
polymers
obtained
results
show
prospects
use
structures
pillar[
n
]arenes
customization
units
controlling
self‐assembly.

Using
supramolecular
host
molecules
to
hinder
bacterial
quorum
sensing
(QS)
is
a
potential
approach
in
circumventing
antimicrobial
resistance
(AMR).
The
emergent
family
of
pillararenes
offers
promising
candidates
for
binding
homoserine
lactones
(HSLs),
signaling
used
by
gram-negative
species,
including
WHO
critical-priority
antibiotic-resistant
bacteria.
Here,
we
compare
seven
cationic
against
four
HSLs,
from
(supra)molecular
interactions
biological
assays.
Complexation,
characterized
dye
displacement
assay
and
NMR
spectroscopy,
complemented
all-atom
molecular
dynamics
(MD)
simulations,
was
compared
effects
systems,
studied
using
HSL
reporter
system
as
well
biofilm
pyocyanin
assays
models
QS-mediated
virulence.
improves
approximately
10-fold
versus
previous
reports
with
hydroxyl-functionalized
pillararene,
deeper-cavity
marked
preference
the
longest-tailed

Successful
capture
directly
reflected
impaired
formation
