Acute kidney injury (AKI) is a global health problem, given its substantial morbidity and mortality rates. A better understanding of the mechanisms factors contributing to AKI has potential guide interventions aimed at mitigating risk subsequent unfavorable outcomes. Endoplasmic reticulum stress (ERS) an intrinsic protective mechanism against external stressors. ERS occurs when endoplasmic (ER) cannot deal with accumulated misfolded proteins completely. ABSTRACT Acute kidney injury (AKI) in diabetic conditions often advances to chronic disease (CKD), exacerbated by ischemia–reperfusion (IRI) through pathomechanisms such as endoplasmic reticulum (ER) stress and inflammation. Currently, available treatment options for AKI are not uniformly effective, highlighting the need novel interventions. This study aimed examine renoprotective effects of hinokitiol, a natural tropolone compound, against with its capability decrease ER inflammation, along apoptosis. involved NRK‐52E cells grown in‐vitro under high‐glucose subjected 10 mM sodium azide elicit hypoxia/reperfusion (HRI). The expression key markers like binding immunoglobulin protein (BiP), R/PKR‐like kinase (PERK), eukaryotic initiation factor‐2 (eIF2α) well inflammatory proteins was markedly diminished hinokitiol pretreatment (50 μM). Hinokitiol further reduced apoptosis cells. It is widely acknowledged that cisplatin-induced nephrotoxicity hinders its efficacy during clinical therapy. Effective pharmaceutical interventions for acute kidney injury (Cis-AKI) are currently lacking. Prior studies have implicated the chemokine CX3CL1 in development of lipopolysaccharide-induced AKI; however, specific role Cis-AKI remains uncertain. This research aimed to comprehensively characterize therapeutic impact and mechanism inhibition on Cis-AKI.This study employed an vivo mouse model vitro cisplatin-treated podocytes. Kidney pathological changes were assessed using hematoxylin-eosin (HE) Periodic-Schiff (PAS) staining. Transcriptome tissue post-cisplatin treatment analyzed through RNA sequencing (RNA-seq) datasets. Evaluation parameters included expression inflammatory markers, intracellular free iron levels, ferroptosis-related proteins-solute carrier family 7 member 11 (SLC7A11/XCT) glutathione peroxidase 4 (GPX4)-as well as lipid peroxidation markers mitochondrial function proteins. In clinical practice, cisplatin is the most commonly used chemotherapy drug to treat a range of malignancies. Severe ROS-regulated nephrotoxicity, however, restricts its applicability. Currently, main mechanisms leading cisplatin-induced nephrotoxicity in settings involve hydration or diuresis. However, not all patients can be treated with massive diuretics. Therefore, it crucial develop treatment modality that effectively reduce through foodborne route. Selenium has been reported have strong antioxidant as well anticancer effects when administered spore oil. Herein, we established cellular and animal models synthesized oil-functionalized nano-selenium (GLSO@SeNPs). We found GLSO@SeNPs inhibit mitochondrial apoptotic pathway by maintaining oxidative homeostasis regulating related signaling pathways (the MAPK, caspase, AKT pathways). vivo, could improve renal impairment, tissues thus inhibiting process injury. addition, were converted into selenocysteine (SeCys2), which may exert protective effects. Oxaliplatin (OXA) is recognized as a first-line drug for gastric cancer. However, low accumulation of the OXA in target site and development resistance directly led to treatment failure. In present study, an ultrasonic extraction method Atractylodes chinensis (DC.) Koidz. polysaccharides (AKUs) combination with vitro were studied. Results showed that when pH level was 11, ultrasound power at 450 W, solid–liquid ratio 1:20, 30 min, yield AKUs significantly increased 13.20 ± 0.35%. The molecular weights ranged from 7.21 185.94 kDa, its monosaccharides mainly composed arabinose (Ara), galactose (Gal), glucose (Glu) 58.36, 16.90, 15.49%, respectively. Cell experiments that, compared alone (2 μg/mL, inhibition rate 18%), had significant synergistic inhibitory effect on MKN45 proliferation, which 33, 41, 45% increasing concentrations (5–50 μg/mL), respectively, representing 2.5-fold inhibition. Inductively coupled plasma-mass spectrometry (ICP-MS) determination confirmed intracellular uptake by 29%, alone. We first demonstrated combined cancer cells mediated reducing expression efflux proteins (MRP1 MRP2) ingested protein (OCT2).
