With the goal of impacting patient quality life and outcomes, sensor science offers significant potential to revolutionize healthcare by providing advances in detection molecular biomarkers for personalized clinical technologies. The community has achieved technical advancements that can impact diagnostics, health monitoring, disease treatment; however, many innovations remain confined laboratory, failing bridge translational gap between research real-world applications. This perspective presents a new direction community, where development centers on needs experiences primary beneficiaries: patients. We provide guidelines resources researchers engage with patients early continuously throughout process inform specifications better align technologies needs, improving their adoption impact. also present examples implementing patient-centered approach planning engagement research. In design impactful sensors patients, must expand focus beyond embrace approach, which will likely lead opportunities collaboration evolution community.

Introduction Senecavirus A (SVA), an emerging vesicular pathogen, is responsible for porcine idiopathic disease (PIVD). This closely associated with and acute neonatal piglet mortality, presenting a substantial threat to the global swine industry. At present, absence of effective drugs or vaccines treating makes accurate diagnosis SVA paramount importance prevention control disease. Methods In this study, we combined reverse transcription loop-mediated isothermal amplification (RT-LAMP) Clustered Regularly Interspaced Short Palindromic Repeats CRISPR-associated protein12a (CRISPR/Cas12a) using dual-labelled fluorescence quencher fluorescent biotin single-stranded DNA reporter molecule develop two rapid, reliable, portable visual assays: RT-LAMP-Cas12a-FQ RT-LAMP-Cas12a-FB. Results The methods exhibited comparable detection limits, 9.6 copies/μL achieved in 40 45 minutes, respectively. They did not cross-react non-target nucleic acids extracted from other related viruses showed high specificity RNA detection. Furthermore, demonstrated satisfactory performance detecting 69 adventitious samples, no significant differences that quantitative polymerase chain reaction (RT-qPCR). Discussion summary, RT-LAMP-Cas12a-FB developed are promising early routine surveillance resource-poor areas.

DNAzyme-based biosensors have emerged as a promising tool for ensuring food safety due to their high sensitivity, specificity, and potential rapid, cost-effective detection of hazardous substances. These leverage DNAzymes-catalytically active DNA molecules-to detect range contaminants, including metal ions, fungal toxins, pesticides, pathogens. While show significant advantages over conventional techniques, challenges such nuclease degradation, interference from complex sample matrices, the costs associated with DNAzyme synthesis still hinder widespread application. Recent advancements in stability DNAzymes, immobilization strategies, integration nanomaterials are progressively addressing these limitations, enhancing performance reliability sensors. This review highlights structural catalytic characteristics assesses current applications safety, discusses innovative strategies overcome existing challenges. The continuous evolution biosensors, particularly design device integration, holds great promise future role routine, reliable analysis.

CRISPR/Cas-based diagnostics offer unparalleled specificity, but their reliance on fluorescently labeled probes and complex nucleic acid extraction limits field applicability. To tackle this problem, we have developed a label-free, equipment-free platform integrating FTA card-based extraction, CRISPR/Cas12a, pre-folded G-quadruplex (G4)–Thioflavin T (ThT) signal reporter. This system eliminates costly fluorescent labeling by leveraging G4-ThT structural binding for visible fluorescence output, while cards streamline isolation without centrifugation. Achieving limit of detection (LOD) to 101 CFU/mL Escherichia coli O157:H7 in spiked food samples, the demonstrated 100% concordance with qPCR standard probe-based CRISPR/Cas12a system. Its simplicity, minimal equipment (portable heating/imaging), cost-effectiveness make it revolutionary tool detecting foodborne pathogens resource-limited environments.
