Background/Objectives:
Protein
aggregation,
particularly
the
aggregation
of
antibody-based
drugs,
has
long
been
a
significant
challenge
in
downstream
processes
and
formulation.
While
inhibitory
effects
excipients
on
have
extensively
studied
using
early
experimental
characterization
methods,
complete
formulation
research
requires
amounts
antibodies
time,
resulting
high
costs.
Methods:
This
study
proposed
quick
small-scale
position-restrained
simulation
method
which
elucidated
mechanism
reversible
self-association
(RSA)
influence
RSA
under
different
conditions.
We
also
validated
rationality
rapid
simulations
through
long-term
(>1
μs)
large-scale
(>1,000,000
atoms)
simulations.
Results:
Through
combing
with
simple
stability
characterization,
monomer
residual
content
trend
shown
concentration
changes
after
thermal
incubation
were
found
to
be
similar
those
observed

Abstract
Over

years,
production

eggs
has
increased
tremendously,
with
an
estimated
global
egg
9.7
billion
by
2050.
Further
processing

popularity.
Liquid

innovative
form
replacement,
still
suffer
from
short
shelf‐life
issues,
and
freezing
been
applied
maintain
freshness.
An
undesirable
phenomenon
called
“gelation”
was
found
during
frozen
which
attracted
numerous
scholars
study
its
mechanism
quality
control
methods.
Therefore,
we
comprehensively
reviewed
history
studies
on

mechanism,

we
describe
a
combined
experimental
computational
approach
investigating
effect
protein–excipient
stability,
which
allows
identification
preferential
interaction

in

marketed
therapeutic
IgG1
monoclonal
antibodies
with
identical
Fc
domain
sequences,
trastuzumab
omalizumab,

investigated

mixtures.

studied
all-atom
molecular
dynamics
(MD)
simulations,
show
accumulation
at
specific
antibody
regions.
Preferential
excipient-interaction
particularly
found
charged
aromatic
residues
complementary-determining
regions,
more
pronounced
arginine
contacts
omalizumab
than
trastuzumab.
These
findings
are
line
stabilizing
observed
long-term
storage
stability
study.
Furthermore,
aggregation
