Abstract Dissolved organic matter (DOM) is widespread in uranium (U) hosting aquifer, a key substance affecting CO 2 + O situ leaching (ISL) of U by consuming injected . However, molecular composition and transformation DOM are poorly understood this aquifer during the mining. To address issue, Fourier transform ion cyclotron resonance mass spectrometry was applied to investigate molecules groundwater samples rock collected from mining aquifer. The high proportion labile tyrosine‐like component BIX value monitoring wells (BM) indicated biological origin DOM. CHOS aliphatic were major components water‐soluble (WSOM) U‐bearing rocks, but CHO highly unsaturated structures with oxygen (HUSHO) main BM. ISL led release WSOM oxidation DOM, which accounted for 24.5%–57.4% 39.1%–65.7% expected respectively. Potential transformations (PMTs) results showed that intragroup dominated, remarkable CHON CHONS occurred ISL. positive net PMTs condensed aromatic structure (CAS), (AS), suggested they transformed HUSLO HUSHO. A gradual decrease (DBE−O)/C wa increase NOSC saturated degree state increased This work successfully identified characteristics proposed paradigm pathways
