Circular RNA (circRNA) is a new endogenous transcription product, which has attracted significant attention in biology research.CircRNA comprise exons or introns involved regulation of various mechanisms.These molecules are stable and species-specific, as well cell tissue-specific.Cardiovascular diseases particularly myocardial ischemia ageing-related diseases, pose major health care burden novel treatments biomarkers should be explored.Recent findings indicate that circRNAs implicated biological processes, such glucose metabolism, fatty acid oxidation, mitochondrial biosynthesis, implying they potential targets for treatment.In the present review, functions heart described, with emphasis given on relationship cardiac aging-related diseases.Directions future research also summarized.

The combination of next-generation sequencing, advanced bioinformatics analysis, and molecular research has now established circular RNAs (circRNAs) as a heterogeneous group non-coding RNA that is widely abundantly expressed. CircRNAs are single-stranded RNA, covalently backspliced to form closed loops. Different models back-splicing have been proposed, mechanisms for circRNA function include sequestering microRNAs, direct interaction with proteins, regulation transcription, translation. Exploring the role circRNAs in different disease settings, understanding how they contribute progression promises provide valuable insight into potential novel therapeutic approaches. Here, we review growing number published on heart cardiovascular system summarize implicated disease.

Atherosclerosis can result in multiple cardiovascular diseases. Circular RNAs (CircRNAs) have been reported as significant non-coding atherosclerosis progression. Dysfunction of vascular smooth muscle cells (VSMCs) is involved atherosclerosis. However, up to now, the effect circ_0002984 still unknown. Currently, we aimed investigate function VSMCs incubated by oxidized low-density lipoprotein (ox-LDL). Firstly, our findings indicated that expression levels were significantly up-regulated serum patients and ox-LDL-incubated VSMCs. Loss suppressed VSMC viability, cell cycle distribution migration capacity. Then, carried out ELISA assay determine TNF-α IL-6 levels. The data implied lack obviously repressed ox-LDL-stimulated inflammation. Meanwhile, miR-326-3p, which was predicted a target circ_0002984, down-regulated treated ox-LDL. Additionally, after overexpression VSMCs, decrease miR-326-3p observed. Subsequently, demonstrated vesicle-associated membrane protein 3 (VAMP3). Therefore, hypothesized could modulate VAMP3 through sponging miR-326-3p. Furthermore, confirmed up-regulation rescued overexpressing-mediated effects on VMSC inhibitor-mediated functions reversed knockdown VAMP3. Circular ribonucleic acids (circRNAs) are a type of RNA that originates through back-splicing events from linear primary transcripts. CircRNAs display high structural resistance and tissue specificity. Accurate quantification the circRNA expression level is vital importance to disease diagnosis. Herein, we construct label-free fluorescent biosensor for ultrasensitive analysis circRNAs based on integration target-initiated cascade signal amplification strategy with light-up G-quadruplex. This assay involves only one assistant probe targets circRNA-specific back-splice junction. When present, it hybridizes initiate duplex-specific nuclease (DSN)-catalyzed cyclic cleavage reaction, producing abundant triggers 3′OH termini. Then, terminal deoxynucleotidyl transferase (TdT) catalyzes addition dGTP dATP at 3′-OH termini resultant obtain long G-rich DNA sequences can form efficient G-quadruplex products. The Thioflavin T (ThT) light up G-quadruplex, generating an enhanced fluorescence. may be performed isothermally without involvement any nucleic acid templates, exogenous primers, specific labeled probes. Importantly, this discriminate target one-base mismatched exhibits good performance in human serum. Moreover, accurately detect cancer cells single-cell even differentiate levels tissues healthy persons nonsmall cell lung (NSCLC) patients, promising applications circRNA-related diagnosis therapeutics.
