Perfluorohexane sulfonic acid (PFHxS) is extensively used in waterproof coatings and fire-fighting foams, several studies have found it to be a potential health hazard, but there still unknown about its effects on spermatogenesis. Our results showed that PFHxS-treated mice significant reproductive toxicity, including decrease sperm count motility, the levels of sex hormones (P < 0.05). Concurrently, structural abnormalities are observed sperm, affecting ≈60-75% those group. Additionally, structure blood-testis barrier (BTB) damaged after PFHxS treatment, leading higher expression inflammatory cytokines microenvironment for Moreover, proteins associated with mitochondrial biogenesis, PTEN-induced kinase 1 (PINK1) NADPH oxidase 4 (NOX4), dysregulated testes treatment. Based metabolome data, differential metabolite 3-hydroxybutanoic identified group, which can regulate histone Kac levels, especially H3K4ac H3K9ac. In summary, this study suggest mice, factors disrupt function metabolic profiles hinder progress gene transcription through Kac, ultimately causing dysfunction.

Hypoxic tumors present a significant challenge in cancer therapy due to their ability adaptation low-oxygen environments, which supports tumor survival and resistance treatment. Enhanced mitophagy, the selective degradation of mitochondria by autophagy, is crucial mechanism that helps sustain cellular homeostasis hypoxic tumors. In this study, we develop an azocalix[4]arene-modified supramolecular albumin nanoparticle, co-delivers hydroxychloroquine mitochondria-targeting photosensitizer, designed induce cascaded oxidative stress regulating mitophagy for treatment These nanoparticles are hypoxia-responsive release loaded guest molecules cells. The released disrupts process, thereby increasing further weakening Additionally, upon laser irradiation, photosensitizer generates reactive oxygen species independent oxygen, inducing damage activation. dual action simultaneous spatiotemporal activation flux blockade results enhanced autophagic stress, ultimately driving cell death. Abstract Evolving concepts on Parkinson’s disease (PD) pathology suggest that α-synuclein (aSYN) promote dopaminergic neuron dysfunction and death through accumulating in the mitochondria. However, consequence of mitochondrial aSYN localisation structure bioenergetic functions neuronal cells are poorly understood. Therefore, we investigated deleterious effects mitochondria-targeted differentiated human neurons comparison with wild-type (WT) overexpression corresponding EGFP (enhanced green fluorescent protein)-expressing controls. Mitochondria-targeted enhanced reactive oxygen species (ROS) formation, reduced ATP levels showed severely disrupted function dendritic neural network, preceding death. Transmission electron microscopy illustrated distorted cristae many fragmented mitochondria response to WT-aSYN overexpression, a complete loss massively swollen expressing aSYN. Further, analysis bioenergetics neurons, WT or aSYN, elicited pronounced impairment respiration. In pharmacological compound screening, found pan-caspase inhibitors QVD zVAD-FMK, specific caspase-1 inhibitor significantly prevented aSYN-induced cell addition, caspase preserved network activity overexpressing Overall, our findings indicated therapeutic by inhibition despite aSYN-mediated alterations morphology function.

Anastasis is a natural cell recovery phenomenon that rescues cells from the brink of death. Programmed death such as apoptosis has been traditionally assumed to be an intrinsically irreversible cascade commits rapid and massive demolition. Interestingly, recent studies have demonstrated dying even at late stages generally considered immutable. Here, we examine evidence for anastasis in cultured animals, review findings illuminating potential mechanisms action, discuss challenges studying explore new strategies uncover function regulation anastasis, identification which wide-ranging physiological, pathological therapeutic implications.

Foam cells are lipid-laden macrophages that contribute to the inflammation and tissue damage associated with many chronic inflammatory disorders. Although foam cell biogenesis has been extensively studied in atherosclerosis, how these form during a infectious disease such as tuberculosis is unknown. Here we report that, unlike cholesterol-laden of tuberculous lung lesions accumulate triglycerides. Consequently, varies underlying disease. In vitro mechanistic studies showed triglyceride accumulation human infected Mycobacterium mediated by TNF receptor signaling through downstream activation caspase cascade mammalian target rapamycin complex 1 (mTORC1). These features distinct from known atherogenic establish new paradigm for non-atherogenic formation. Moreover, they reveal novel targets disease-specific pharmacological interventions against maladaptive macrophage responses.
