The implementation of immune checkpoint inhibitors to the oncology clinic signified a new era in cancer treatment. After first indication melanoma, an increasing list additional types are now treated with system targeting antibodies PD-1, PD-L1 and CTLA-4, alleviating inhibition signals on T cells. Recently, we published proof-of-concept results novel inhibitor, NKG2A. This receptor is expressed cytotoxic lymphocytes, including NK cells subsets activated CD8+ Blocking NKG2A unleashed reactivity these effector resulting tumor control multiple mouse models early clinical trial. Monalizumab inhibiting this human beings future trials will have reveal its potency combination other treatment options.

These guidelines are a consensus work of considerable number members the immunology and flow cytometry community. They provide theory key practical aspects enabling immunologists to avoid common errors that often undermine immunological data. Notably, there comprehensive sections all major immune cell types with helpful Tables detailing phenotypes in murine human cells. The latest techniques applications also described, featuring examples data can be generated and, importantly, how analysed. Furthermore, tips, tricks pitfalls avoid, written peer-reviewed by leading experts field, making this an essential research companion.

Abstract Identifying tumor antigen-specific T cells from cancer patients has important implications for immunotherapy diagnostics and therapeutics. Here, we show that CD103 + CD39 tumor-infiltrating CD8 (CD8 TIL) are enriched tumor-reactive both in primary metastatic tumors. This TIL subset is found across six different malignancies displays an exhausted tissue-resident memory phenotype. TILs have a distinct T-cell receptor (TCR) repertoire, with clones expanded the but present at low frequencies periphery. also efficiently kill autologous MHC-class I-dependent manner. Finally, higher of head neck associated better overall survival. Our data thus describe approach detecting will help define mechanisms existing treatments, may lead to future adoptive therapies.

Abstract The NOTCH gene was identified approximately 110 years ago. Classical studies have revealed that signaling is an evolutionarily conserved pathway. receptors undergo three cleavages and translocate into the nucleus to regulate transcription of target genes. deeply participates in development homeostasis multiple tissues organs, aberration which results cancerous noncancerous diseases. However, recent indicate outcomes are changeable highly dependent on context. In terms cancers, can both promote inhibit tumor various types cancer. overall performance NOTCH-targeted therapies clinical trials has failed meet expectations. Additionally, mutation been proposed as a predictive biomarker for immune checkpoint blockade therapy many cancers. Collectively, pathway needs be integrally assessed with new perspectives inspire discoveries applications. this review, we focus classical latest findings related illustrate history, architecture, regulatory mechanisms, contributions physiological development, diseases, therapeutic applications microenvironment cancer immunotherapy also highlighted. We hope review will help not only beginners but experts systematically thoroughly understand

Human T cells coordinate adaptive immunity in diverse anatomic compartments through production of cytokines and effector molecules, but it is unclear how tissue site influences cell persistence function. Here, we use single RNA-sequencing (scRNA-seq) to define the heterogeneity human isolated from lungs, lymph nodes, bone marrow blood, their functional responses following stimulation. Through analysis >50,000 resting activated cells, reveal signatures mucosal lymphoid sites, lineage-specific activation states across all sites including distinct for CD8+ an interferon-response state CD4+ cells. Comparing scRNA-seq profiles tumor-associated our dataset reveals predominant compared within multiple tumor types. Our results therefore establish a high dimensional reference map health analyzing disease.

The discovery of T cells resident in diverse tissues has altered our understanding adaptive immunity to encompass site-specific responses mediated by tissue-adapted memory throughout the body. Here, we discuss key phenotypic, transcriptional, and functional features these tissue-resident (TRM) as established mouse models infection translated humans novel tissue sampling approaches. Integration findings from human studies may hold unlocking potential TRM for promoting preventing infection.
