Over the past decade, Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for definition and interpretation of cell death from morphological, biochemical, functional perspectives. Since field continues to expand novel mechanisms that orchestrate multiple pathways are unveiled, we propose an updated classification subroutines focusing mechanistic essential (as opposed correlative dispensable) aspects process. As provide molecularly oriented definitions terms including intrinsic apoptosis, extrinsic mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic death, NETotic lysosome-dependent autophagy-dependent immunogenic cellular senescence, mitotic catastrophe, discuss utility neologisms refer highly specialized instances these processes. The mission NCCD is a widely accepted nomenclature in support continued development field.

Importance

Osteoarthritis (OA) is the most common joint disease, affecting an estimated more than 240 million people worldwide, including 32 in US. frequent reason for activity limitation adults. This Review focuses on hip and knee OA.

Observations

can involve almost any but typically affects hands, knees, hips, feet. It characterized by pathologic changes cartilage, bone, synovium, ligament, muscle, periarticular fat, leading to dysfunction, pain, stiffness, functional limitation, loss of valued activities, such as walking exercise dancing. Risk factors include age (33% individuals older 75 years have symptomatic radiographic OA), female sex, obesity, genetics, major injury. Persons with OA comorbidities are sedentary those without The reduced physical leads a 20% higher age-adjusted mortality. Several examination findings useful diagnostically, bony enlargement pain elicited internal rotation Radiographic indicators marginal osteophytes space narrowing. cornerstones management exercises, weight if appropriate, education—complemented topical or oral nonsteroidal anti-inflammatory drugs (NSAIDs) contraindications. Intra-articular steroid injections provide short-term relief duloxetine has demonstrated efficacy. Opiates should be avoided. Clinical trials shown promising results compounds that arrest structural progression (eg, cathepsin K inhibitors, Wnt anabolic growth factors) reduce nerve factor inhibitors). advanced symptoms damage candidates total replacement. Racial ethnic disparities persist use outcomes

Conclusions Relevance

Hip highly prevalent disabling. Education, management, complemented NSAIDs (for patients who candidates), corticosteroid injections, several adjunctive medications. For persons damage, replacement effectively relieves pain.

Aging is the major risk factor for cancer, cardiovascular disease, diabetes, and neurodegenerative disorders. Although we are far from understanding biological basis of aging, research suggests that targeting aging process itself could ameliorate many age-related pathologies. Senescence a cellular response characterized by stable growth arrest other phenotypic alterations include proinflammatory secretome. plays roles in normal development, maintains tissue homeostasis, limits tumor progression. However, senescence has also been implicated as cause disease. In this regard, recent experimental evidence shown genetic or pharmacological ablation senescent cells extends life span improves health span. Here, review molecular links between discuss novel therapeutic avenues connection opens.

Cellular senescence is a permanent state of cell cycle arrest that occurs in proliferating cells subjected to different stresses. Senescence is, therefore, cellular defense mechanism prevents the acquire an unnecessary damage. The senescent accompanied by failure re-enter response mitogenic stimuli, enhanced secretory phenotype and resistance death. takes place several tissues during physiological pathological processes such as tissue remodeling, injury, cancer, aging. Although one causative aging it responsible aging-related disorders, can also play positive role. In embryogenesis are required for proper development embryo repair. works potent barrier prevent tumorigenesis. Therefore, identification characterization key features senescence, induction cancer cells, or elimination pharmacological interventions gaining consideration fields research. Here, we describe known cell-autonomous, noncell-autonomous regulators attempt discuss functional role this fundamental process contexts light novel therapeutic targets.
