Tremendous progress has been made since Neuron published our Primer on genetic dissection of neural circuits 10 years ago. Since then, cell-type-specific anatomical, neurophysiological, and perturbation studies have carried out in a multitude invertebrate vertebrate organisms, linking neurons to behavioral functions. New methods allow systematic classification cell types provide access diverse neuronal for connectivity coding during behavior. Here we evaluate key advances over the past decade discuss future directions.

Abstract Metasurface is a recently developed nanophotonics concept to manipulate the properties of light by replacing conventional bulky optical components with ultrathin (more than 10 4 times thinner) flat components. Since first demonstration metasurfaces in 2011, they have attracted tremendous interest consumer optics and electronics industries. Recently, metasurface-empowered novel bioimaging biosensing tools emerged been reported. Given recent advances biomedical engineering, this review article covers state art for technology provides comprehensive interdisciplinary perspective on field. The topics that we covered include chiral imaging, endoscopic coherence tomography, fluorescent super-resolution magnetic resonance quantitative phase sensing antibodies, proteins, DNAs, cells, cancer biomarkers. Future directions are discussed twofold: application-specific bioinspired metasurface devices. The active properties of dendrites can support local nonlinear operations, but previous imaging and electrophysiological measurements have produced conflicting views regarding the prevalence selectivity nonlinearities in vivo. We imaged calcium signals pyramidal cell motor cortex mice performing a tactile decision task. A custom microscope allowed us to image soma up 300 μm contiguous dendrite at 15 Hz, while resolving individual spines. New analysis methods were used estimate frequency spatial scales activity dendritic branches majority transients coincident with global events. However, task-associated spines compartmentalized by branching clustered within over approximately 10 μm. Diverse behavior-related intermingled distributed throughout arbor, potentially supporting large learning capacity neurons.

Whole-brain recordings give us a global perspective of the brain in action. In this study, we describe method using light field microscopy to record near-whole calcium and voltage activity at high speed behaving adult flies. We first obtained maps for various stimuli behaviors. Notably, found that increased on scale when fly walked but not it groomed. This increase with walking was particularly strong dopamine neurons. Second, extracted spatially distinct sources as well their time series principal component analysis independent analysis. The characteristic shapes matched anatomy subneuropil regions and, some cases, specific neuron type. Brain structures responded odor were consistent previous reports, confirming new technique's validity. also observed previously uncharacterized behavior-related patterns spontaneous activity.
