Sleep plays an important role in maintaining neuronal circuitry, signalling and helps maintain overall health wellbeing. deprivation (SD) disturbs the circadian physiology exerts a negative impact on brain behavioural functions. SD impairs cellular clearance of misfolded neurotoxin proteins like α-synuclein, amyloid-β, tau which are involved major neurodegenerative diseases Alzheimer's disease Parkinson's disease. In addition, is also shown to affect glymphatic system, glial-dependent metabolic waste pathway, causing accumulation faulty synaptic compartments resulting cognitive decline. Also, affects immunological redox system neuroinflammation oxidative stress. Hence, it understand molecular biochemical alterations that causative factors leading these pathophysiological effects system. This review attempt this direction. It provides up-to-date information key processes, pathways, negatively affected by become reasons for neurological disorders over prolonged period time, if left unattended.

Background: Over recent decades, a growing body of evidence has emerged linking the composition gut microbiota to sleep regulation. Interestingly, prevalence disorders is commonly related cardiometabolic comorbidities such as diabetes, impaired lipid metabolism, and metabolic syndrome (MetS). In this complex scenario, role gut–brain axis main communicating pathway between regulation pathways in brain reveals some common host–microbial biomarkers both disturbances MetS. Further evidence has accumulated suggesting role in cancer initiation progression (primarily breast cancer). patients with survivors frequently experience sleep, manifesting as insomnia, circadian misalignment, hypersomnia, somnolence hot flushes, nightmares.
