Local translation regulates the axonal proteome, playing an important role in neuronal wiring and axon maintenance. How mRNAs are localized to specific subcellular sites for translation, however, is not understood. Here we report that RNA granules associate with endosomes along axons of retinal ganglion cells. RNA-bearing Rab7a late also ribosomes, real-time imaging reveals they local protein synthesis. We show often pause on mitochondria encoding proteins mitochondrial function translated endosomes. Disruption mutants, including those associated Charcot-Marie-Tooth type 2B neuropathy, markedly decreases synthesis, impairs function, compromises viability. Alzheimer's disease (AD) is a progressive neurodegenerative characterized by memory loss and multiple cognitive impairments. Several decades of intense research have revealed that cellular changes are implicated in the development progression AD, including mitochondrial damage, synaptic dysfunction, amyloid beta (Aβ) formation accumulation, hyperphosphorylated tau (P-Tau) deregulated microRNAs, neuronal patients with AD.
