Pectin is a heteropolysaccharide that acts as an intestinal immunomodulator, promoting development and regulating flora in the gut. However, relevant mechanisms remain obscure. In this study, pigs were fed corn-soybean meal-based diet supplemented with either 5% microcrystalline cellulose (MCC) or pectin for 3 weeks, to investigate metabolites anti-inflammatory properties of jejunum.The results showed dietary supplementation improved integrity (Claudin-1, Occludin) inflammatory response [interleukin (IL)-10], expression proinflammatory cytokines (IL-1β, IL-6, IL-8, TNF-α) was down-regulated jejunum. Moreover, altered jejunal microbiome tryptophan-related piglets. specifically increased abundance Lactococcus, Enterococcus, microbiota-derived (skatole (ST), 3-indoleacetic acid (IAA), 3-indolepropionic (IPA), 5-hydroxyindole-3-acetic (HIAA), tryptamine (Tpm)), which activated aryl hydrocarbon receptor (AhR) pathway. AhR activation modulates IL-22 its downstream pathways. Correlation analysis revealed potential relationship between morphology, gene expression, cytokine levels.In conclusion, these indicated inhibits by enhancing AhR-IL22-signal transducer activator transcription signaling pathway, through tryptophan metabolites.
