Introduction The complex and unresolved pathogenesis of schizophrenia has posed significant challenges to its diagnosis treatment. While recent research established a clear association between immune function schizophrenia, the causal relationship two remains elusive. Methods We employed bidirectional two-sample Mendelian randomization approach investigate 731 cell traits by utilizing public GWAS data. further validated six types white measures. Results found overall effects on were significantly higher than reverse ones (0.011 ± 0.049 vs 0.001 0.016, p < 0.001), implying that disease may lead an increase in cells itself. also identified four risk schizophrenia: CD11c+ monocyte %monocyte (odds ratio (OR): 1.06, 95% confidence interval (CI): 1.03~1.09, FDR = 0.027), CD62L- (OR:1.06, CI: CD25 IgD+ CD38- naive B (OR:1.03, CI:1.01~1.06, 0.042), CD86 (OR 1.04, 0.042). However, we did not detect any traits. Using blood data, increases lymphocyte counts 95%CI: 1.01-1.04, 0.007), total (OR:1.02, 0.021) 1.00-1.03, 0.034). nominally associated with (OR:1.08,95%CI:1.01-1.16, P=0.019). Discussion Our study system is complex, enhancing our understanding role regulation development this disorder. These findings offer new insights for exploring diagnostic therapeutic options schizophrenia.

Abstract Background There is an association between migraine and dementia, however, their causal relationship remains unclear. This study employed bidirectional two-sample Mendelian randomization (MR) to investigate the potential dementia its subtypes: Alzheimer’s disease (AD), vascular (VaD), frontotemporal (FTD), with Lewy bodies (DLB). Methods Summary-level statistics data were obtained from publicly available genome-wide studies (GWAS) for both five types of dementia. Single nucleotide polymorphisms (SNPs) associated each subtype selected. MR analysis was conducted using inverse variance weighting (IVW) weighted median (WM) methods. Sensitivity analyses included Cochran’s Q test, pleiotropy residual sum outlier (MR-PRESSO) analysis, intercept MR-Egger, leave-one-out analysis. Results Migraine showed a significant AD VaD, whereas no observed all-cause FTD, or DLB. may be risk factor (odds ratio [OR]: 1.09; 95% confidence interval [CI]: 0.02–0.14; P = 0.007), while VaD (OR: 1.04; CI: 0.02–0.06; 7.760E-5). demonstrated robustness our findings. Conclusion Our suggest that have relationships VaD. AD, migraine. contributes unraveling comprehensive genetic associations various findings will enhance academic understanding comorbidity

Abstract The connection between periodontal disease (PD) and schizophrenia (SCZ) has been reported in observational studies, but it remains unclear. This research aims to examine the bidirectional causal impacts PD SCZ. FinnGen consortium supplied summarized data on for 346,731 individuals (87,497 cases 259,234 controls) of Finnish ancestry, information SCZ was acquired from OpenGWAS repository, encompassing 127,906 (52,017 75,889 European ancestry. Next, we conducted Mendelian randomization (MR) analysis establish a inference relationship inverse variance weighted (IVW) method utilized as primary analysis. Additionally, some sensitivity analyses were verify stability results. IVW results indicated no impact (IVW OR = 1.10, 95% CI 0.97–1.24, P 0.14). Nevertheless, identified through reverse MR 1.03, 1.01–1.05, 0.002). findings MR-Egger, median, simple mode, mode approaches aligned with outcomes method. Based analyses, horizontal pleiotropy is unlikely distort estimates. study presented initial proof genetic PD, albeit minimal impact. Further exploration needed gain deeper understanding this relationship. Furthermore, identified.

ABSTRACT Background Observational studies have suggested that obsessive–compulsive disorder (OCD) may be associated with Alzheimer's disease (AD). However, whether OCD is a causal risk factor for AD remains unclear. This study aimed to assess the effect of on by performing two‐sample Mendelian randomization (MR) analysis. Methods Genome‐wide association summary statistics were obtained OCD, comprising 2688 cases and 7037 controls, as well AD, including 21,982 41,944 controls from Kunkle et al.’s study, 39,918 358,140 Wightman study. On basis two diverse thresholds, OCD‐associated genetic variants screened instrumental variables (IVs) subsequent MR analyses. Inverse variance weighed was primary method. MR‐Egger, weighted median, mode used supplementary methods. Various sensitivity tests assessed reliability results. Results strict IV selecting inverse‐variance (IVW) identified significant associations between liability increased in different sources ((i) al.: odds ratio [OR] = 1.070, 95% confidence interval [CI]: 1.015–1.127, p 0.012; (ii) al. (iii) OR 1.051, CI: 1.014–1.090, 0.007). Three other methods yielded similar results IVWs (OR > 1). Furthermore, all replicated analyses based lenient thresholds. The indicated stable not affected horizontal pleiotropy. Conclusions comprehensive suggests AD. Early intervention patients beneficial preventing future progression.
