Epigenetic and metabolic reprogramming alterations are two important features of tumors, their reversible, spatial, temporal regulation is a distinctive hallmark carcinogenesis. Epigenetics, which focuses on gene regulatory mechanisms beyond the DNA sequence, new entry point for tumor therapy. Moreover, drives hepatocellular carcinoma (HCC) initiation progression, highlighting significance metabolism in this disease. Exploring inter-regulatory relationship between epigenetic modification has become one hot directions current research. As viral etiologies have given way to dysfunction-associated steatotic liver disease (MASLD)-induced HCC, it urgent that complex molecular pathways linking them hepatocarcinogenesis be explored. However, how aberrant crosstalk modifications affects MASLD-induced HCC lacks comprehensive understanding. A better understanding linkages necessary improve treatment strategies. For reason, review examines interwoven landscape carcinogenesis context focusing regulating development interactions while also updating advances modification-based therapeutic drugs HCC.

Glycolysis is a vital metabolic biological process in tumor progression and immune modulation. This study comprehensively investigated the roles of glycolysis pan-cancer, especially bladder cancer. Exploration 34 single-cell RNA sequencing (scRNA-seq) cohorts, eight ICI-treated bulk RNA-seq TCGA pan-cancer cohorts uncovered Glycolysis.Sig which strongly correlated with immunotherapy response demonstrated excellent predictive performance prognosis response. Hub-Glycolysis.Sig exhibited varying interactions microenvironment based on cancer type. In cancer, higher risk scores poorer prognosis, distinct infiltration characteristics between subtypes. scRNA-seq revealed high levels epithelial cells. COPB2 was highly expressed promoting cell proliferation, migration, glycolytic activity vitro vivo. Our large-scale data analysis confirmed negative correlation outcomes, identifying as novel biomarker. provides clinical insights for therapy strategies, while other potential therapeutic targets facilitate personalized treatment.

Hepatocellular carcinoma (HCC) is a malignant tumour with high prevalence and mortality rate worldwide. Metabolic reprogramming of cancer cells may be major factor in the process this disease. Glucose transporter proteins (GLUTs) are members facilitator superfamily membrane transporters, playing pivotal role metabolic progression HCC. This review discusses advances study GLUTs HCC, including expression patterns, functions possibilities GLUTs. In levels closely associated aggressiveness, prognosis. A series inhibitors have been demonstrated efficacy inhibiting HCC cell growth glucose uptake vitro vivo models. These offer novel approach to treatment by reducing metabolism cells, thereby impeding growth, concurrently enhancing sensitivity chemotherapeutic agents. reminds us urgent need elucidate GLUTs’ roles determine most effective ways translate these findings into clinical practice.

Cells universally employ an efficiency-driven metabolic switch mechanism during nutritional changes, growth, and differentiation, transitioning from oxidative phosphorylation (OXPHOS) to glycolysis ensure survival under hypoxic conditions or high energy demands. In cerebral ischemia, inadequate blood supply causes oxygen deprivation, prompting brain cells initiate glycolytic reprogramming meet urgent needs. While this adaptation is a temporary solution, it may lead lactic acidosis, aggravated inflammation, increased free radical production. Prolonged reperfusion with sustained can exacerbate cell damage, potentially causing irreversible harm. This review systematically examines the dynamic changes in glucose transport mechanisms roles of immediate, early, intermediate, late responder cells, along their regulatory factors, reprogramming. Using temporal analysis framework based on body's natural response sequence pathological events, we elucidate how at different stages collaborate address metabolism conditions. Reversing inhibiting improve processes ischemic stroke, offering potential therapeutic benefits.

Acute myeloid leukemia (AML), an aggressive malignancy of hematopoietic stem cells, is characterized by the blockade cell differentiation, uncontrolled proliferation, and expansion that impairs healthy hematopoiesis results in pancytopenia susceptibility to infections. Several genetic chromosomal aberrations play a role AML influence patient outcomes. TP53 key tumor suppressor gene involved variety features, such as cell-cycle regulation, genome stability, stem-cell homeostasis, apoptosis, metabolism, senescence, repair DNA damage response cellular stress. In AML, alterations occur 5%-12% de novo cases. These mutations form important molecular subgroup, patients with these have worst prognosis shortest overall survival among even when treated chemotherapy allogeneic transplant. The frequency TP53-mutations increases relapsed recurrent associated chemoresistance. Progress genetics biology has brought novel therapies, however, clinical benefit agents for whose disease driven remains largely unexplored. This review focuses on characteristics TP53-mutated disease; impact selected hallmarks leukemia, particularly metabolic rewiring immune evasion, importance mutations; current progress development preclinical therapeutic strategies treat disease.

Considering that muscle pH is one of the crucial factors for pork quality, influenced by characteristics such as rate and extent its decline, pig breeds, glycolytic enzymes. To understand mechanisms related to energy metabolism muscle-to-meat conversion, this study aimed investigate existence distinct gene expressions may explain through analysis characterize it in different breeds. For purpose, 39 animals from four genetic groups were used: Piau (PP), Large White (LW), crossbreeds Piau-Large (PL) Duroc-Large (DL). Muscle measurements collected at eleven times post-slaughter (0, 0.75, 1, 2, 3, 4, 5, 6, 7, 8, 24 hours), which used repeated measures model over time, including group, time-genetic group interaction a fixed effect, followed mean analysis, where some significant differences between observed (p<0.05). The decline was conducted using exponential model. expression Longissimus dorsi samples utilized, selected based on total RNA concentration sample. Ten genes post-mortem selected, with Additionally, pattern observed, PP PL showed higher relative contributing better quality. Furthermore, local demonstrated predominantly additive behavior, retaining favorable characteristics, how best values slower rate. From these results, concluded altering selection, specifically use has potential modulate levels and, consequently, optimize final quality indices.
