In the large poultry industry, where farmed chickens are fed at high density, prevalence of pathogens and repeated vaccinations induce immune stress, which can significantly decrease production performance increase mortality. This study was designed to shed light on molecular mechanisms metabolic pathways involved in stress through an in-depth analysis transcriptomic metabolomic changes jejunum samples from broilers. Two groups were established for experiment: a control group LPS group. received intraperitoneal injection solution dose 250 μg per kg 12, 14, 33, 35 days age, whereas sterile saline injection. The severity assessed using Disease Activity Index. A jejunal section collected measure intestinal villus structure (villus length crypt depth). RNA sequencing metabolomics data conducted reveal differentially expressed genes metabolites. results showed that DAI index increased height/crypt depth decreased total 96 672 accumulating metabolites detected by compared comprehensive 23 enriched appetite, nutrient absorption, energy substance metabolism disorders ferroptosis play important role Our findings provide deeper understanding responses broilers LPS-induced suggesting potential targets therapeutic strategies improve broiler chickens.

Background: Parkinson's disease (PD) is a neurodegenerative disorder characterized by the involvement of ferroptosis in its pathological mechanism. In this study, effects and mechanism BRCA1-associated protein 1 (BAP1) on neuronal PD were evaluated. Methods: A mouse model was constructed injecting mice with MPTP. Nissl staining, immunohistochemistry, immunofluorescence, Prussian blue staining evaluated histopathology iron distribution. Calcium oxalate (CaOx) stones are among the most common types of kidney and associated with renal tubular damage, interstitial fibrosis, chronic disease. The mechanism CaOx crystal-induced fibrosis remains unknown. Ferroptosis, a type regulated cell death, is characterised by iron-dependent lipid peroxidation, tumour suppressor p53 key regulator ferroptosis. In present study, our results demonstrated that ferroptosis was significantly activated in patients nephrolithiasis hyperoxaluric mice as well verified protective effects inhibition on fibrosis. Moreover, single-cell sequencing database, RNA-sequencing, western blot analysis revealed expression increased disease oxalate-stimulated human epithelial line, HK-2. Additionally, acetylation enhanced stimulation HK-2 cells. Mechanistically, we found induction deacetylation, owing to either SRT1720-induced activation deacetylase sirtuin 1 or triple mutation p53, inhibited alleviated caused crystals. We conclude one critical mechanisms contributing pharmacological via 1-mediated deacetylation may be potential target for preventing nephrolithiasis.
