Endometrial polyps and uterine leiomyomas are common gynecological conditions that significantly affect women's health. Recent studies have begun to explore potential biomarkers could assist in the early diagnosis understanding of pathophysiology these conditions. The role of fat mass and obesity-associated protein (FTO), an N6-methyladenosine (m6A) demethylase, in non-small cell lung cancer (NSCLC) has recently received widespread attention. Necroptosis, a novel type of programmed cell death, is intricately associated with inflammatory response. Currently, most studies focus on the activation necroptosis, while mechanisms underlying negative regulation necroptosis remain poorly understood. The effects sestrin2 (SESN2) overexpression or knockdown were assessed in TNFα/Smac-mimetic/Z-VAD-FMK (T/S/Z)-induced model and palmitic acid (PA)-induced lipotoxicity model. Western-blot, co-Immunoprecipitation, Glutathione S-transferase pull-down, confocal assays employed to explore regulatory including protein-protein interactions post-translational modification. Furthermore, we used GSK'872, specific inhibitor receptor-interacting serine/threonine-protein kinase (RIPK) 3, evaluate relationship between SESN2-related alterations RIPK3-mediated T/S/Z-induced model, PA-induced high-fat high-cholesterol diet (HFHCD)-induced non-alcoholic steatohepatitis Our findings revealed that SESN2 was upregulated under conditions induce functioned as regulator necroptosis.
