Ferroptosis is a novel form of cell death distinct from traditional mechanisms, characterized by the accumulation iron ions and production lipid peroxides. It not only affects survival tumor cells but also closely linked to changes in microenvironment. Lung cancer one leading malignancies worldwide terms incidence mortality, its complex biological mechanisms resistance make treatment challenging. Recent studies have shown that ferroptosis plays key role onset progression lung cancer, with intricate regulatory influencing development response therapy. As research into deepens, related molecular pathways, such as glutamate metabolism, antioxidant defense, been gradually revealed. However, clinical practice, ferroptosis-based therapeutic strategies for are still their early stages. Challenges remain, including incomplete understanding specific ferroptosis, insufficient on factors, limited insight interactions within Therefore, effective modulation enhance remains an urgent issue. This review summarizes analyzes factors interaction microenvironment, further explores potential targeting ferroptosis. By synthesizing latest research, this paper aims provide new perspectives directions treatment, goal advancing applications.

Abstract Ferroptosis has been characterised by disruption of the cell membrane through iron‐related lipid peroxidation. However, regulation iron homeostasis in lung cancer cells that are resistant to epidermal growth factor receptor tyrosine kinase inhibitors (EGFR‐TKIs) remains unclear. Transcriptome analysis identified a significant downregulation apoptosis‐associated ( AATK ) mRNA expression gefitinib‐resistant PC9 (PC9‐GR) cells, which were found be more susceptible ferroptosis inducers. An in‐depth publicly available datasets revealed was associated with lymph node metastasis and poor prognosis patients adenocarcinoma. Knockdown AATK‐sensitised PC9, HCC827, H441 inducer RSL3, whereas ectopic reduced RSL3‐induced death PC9‐GR HCC827‐GR cells. Compared exhibited higher transferrin uptake, endosome recycling rate, increased intracellular levels. Blocking transport Mechanistic studies showed localised both early endosomes. facilitated elevated ferrous (Fe 2+ levels Conversely, delayed Fe Inhibition downregulation‐induced accumulation decreased ferroptosis. Taken together, our study indicates contributes accumulation, leading an susceptibility EGFR‐TKI‐resistant © 2025 The Pathological Society Great Britain Ireland.

Objectives This study employs bibliometric analysis to track evolution and identify trends of key topics in ferroptosis within the context lung cancer. By identifying emerging research areas, our aim is provide valuable insights directions for researchers this field. Methods Relevant papers reviews on cancer were retrieved from Web Science Core Collection database 5 February 2024. Bibliometric was conducted using CiteSpace 6.2.R3, VOSviewer 1.6.20, R 4.3.0, Microsoft Excel 2019. Results From 2015 2020, publications related sparse but showed a steady increase. Post-2020, there has been significant surge, with 6.4-fold increase observed by 2023. Overall, authors 4,033 institutions across 42 countries/regions contributed 606 published 262 academic journals. China emerged as leading contributor, while United States maintained dominance. Lifang Ma most prolific author, DIXON SJ, YANG WS, STOCKWELL BR being frequently co-cited. Effective communication collaboration among scholars are lacking. Key journals include Frontiers Pharmacology publication output, Nature Cell citation frequency. Research focuses molecular mechanisms ferroptosis, including endoplasmic reticulum stress, tumor microenvironment, autophagy. Therapeutic targets like GPX4, SLC7A11, P53, FSP1, Nrf2, LSH, STYK1/NOK, ACSL4 prominent. Traditional Chinese medicine also shows clinical value research. Conclusion Ferroptosis, promising avenue applications cancer, continues undergo rapid development. The iron death will remain critical focus future.
