We recently reported on the development of a unique cancer-targeting peptide called NAF-1 44-67 (derived from CISD2/NAF-1). selectively permeates plasma membrane (PM) cancer cells, but not healthy causing activation apoptotic and ferroptotic cell death pathways specifically in cells. also targets shrinks human breast ovarian tumors xenograft mice model system without any apparent side effects. Although specific permeation through PMs was studied, its killing effects validated vitro vivo , little is known about how exerts biological activity once it enters Here, we report that CISD2/NAF-1 protein cells disrupts homodimeric structure. further reveal derived same domain CISD1 (mitoNEET; mNT 19-42 ) protein, close family member to CISD2, has no towards dimers (at two different orientations) have higher anticancer compared monomeric . Our findings shed new light bring closer becoming potential drug.
