The circadian clock regulates various physiological processes in mammals. core gene Bmal1 is crucial for maintaining the oscillations of system by controlling rhythmic expression numerous clock-controlled genes. To explore transcriptional changes associated with deletion liver tissues, we collected tissues from global and liver-specific knockout mice, along their respective control groups, at two time points (CT2 CT14) used them transcriptome sequencing analysis. Genotyping, locomotor activity analysis, comprehensive quality analyses, including base scores, GC content, mapping rates, confirmed high data. Differential analysis RT-qPCR validation reliability validity dataset. These data offer a valuable resource researchers investigating role BMAL1 physiology, pathology, broader field biology.

Breast cancer (BC) is one of the most common and fatal malignancies among women worldwide. Circadian rhythms have emerged in recent studies as being involved pathogenesis breast cancer. In this paper, we reviewed molecular mechanisms by which dysregulation circadian genes impacts development BC, focusing on critical clock genes, brain muscle ARNT-like protein 1 (BMAL1) locomotor output cycles kaput (CLOCK). We discussed how rhythm disruption (CRD) changes tumor microenvironment (TME), immune responses, inflammation, angiogenesis. The CRD compromises surveillance features activities effectors, including CD8+ T cells tumor-associated macrophages, that are important an effective anti-tumor response. Meanwhile, review, discuss bidirectional interactions: age rhythms, aging further increases risk through reduced vasoactive intestinal polypeptide (VIP), affecting suprachiasmatic nucleus (SCN) synchronization, ability to repair damaged DNA, weakened immunity. These complex interplays open new avenues toward targeted therapies combination drugs with chronotherapy potentiate response while reducing progression for better outcomes. Abstract Cancer immunotherapies, including immune checkpoint blockade (ICB), have marked a significant breakthrough in cancer treatment but their clinical efficacy is limited immune-resistant tumors. Previously, we found that immunotherapy-mediated selection enriches tumors with both tumor-intrinsic and -extrinsic refractory phenotypes via the transcriptional induction of HDAC1 by NANOG. Here, identify CRY1 as critical target NANOG stabilizes Cyclin A MCL1 to promote stem cell-like property resistance cytotoxic T cell-mediated killing NANOGhigh tumor cells through HDAC1-mediated epigenetic silencing APC3 TRIM17. Additionally, downregulates CXCL10 repression, thereby suppressing cell infiltration. Importantly, inhibition synergizes PD-1 adoptive transfer reducing growth converting into immune-sensitive Collectively, these findings highlight mediator NANOG/HDAC1 axis multiple properties suggest potential therapeutic target.

Healthcare workers (HCWs) are frequently exposed to a variety of chemical agents, which can result in the development allergic or irritant contact dermatitis. The present study aimed assess prevalence skin sensitization among HCWs who presented with symptoms dermatitis, considering both occupational and non-occupational risk factors. population comprised 127 attended routine health surveillance at Tor Vergata Teaching Hospital Rome between November 2023 May 2024. A structured dermatitis questionnaire patch testing were administered participants. Demographic lifestyle data, including information on occupation, night shift work, smoking habits, body mass index (BMI), collected. Patch test positivity was observed 31.5% participants, most common clinical presentation being erythematous-desquamative significantly higher likelihood nurses (57.1%), particularly for nickel sensitization, compared other groups. trend towards an association work observed, although this did not reach statistical significance. No significant associations found ages over 35 years, sex, BMI. These findings highlight elevated nurses, emphasizing need targeted interventions, exposure reduction strategies protective measures, mitigate hazards healthcare settings.
