Abstract Mast cells play a central role in allergic reactions, acting as key effector that initiate and amplify the inflammatory response. In this study, we demonstrate phosphatase of regenerating liver 2 (PRL2) functions negative regulator FcεRI-mediated mast cell activation. PRL2-deficient myeloid cells, PRL2 conditional knockout mice developed more severe passive systemic anaphylaxis (PSA). Although deficiency does not impact development, absence activation is enhanced. presence IgE expression downregulated, leading to modulation cellular PI3K signaling pathway upregulated, resulting increased calcium influx.
