The advent of highthroughput ‘omics’ technologies has improved our knowledge gut microbiome in human health and disease, including Alzheimer’s disease (AD), a neurodegenerative disorder. Frequent bidirectional communications mutual regulation exist between the gastrointestinal tract central nervous system through gut-brain axis. A large body research reported close association microbiota AD development, restoring healthy may curb or even improve symptoms progression. Thus, modulation become novel paradigm for clinical management AD, emerging effort focused on developing potential strategies preventing and/or treating disease. In this review, we provide an overview connection causal relationship dysbiosis mechanisms driving progression, successes challenges implementing available microbiome-targeted therapies (including probiotics, prebiotics, synbiotics, postbiotics, fecal transplantation) preventive therapeutic preclinical intervention studies AD. Finally, discuss future directions field.

Abstract Yeasts, with a history of approximately 5,000 years in food and medicinal applications, play vital role various industries. The advantages these microorganisms include probiotic effects, phytate biodegradation, mycotoxin breakdown, adsorption capabilities. Traditionally, research on probiotics has primarily focused lactic acid bacteria bifidobacteria; however, there is growing global interest incorporating yeast as supplement. Notably, species such Saccharomyces , Candida Debaryomyces Yarrowia Kluyveromyces have been identified for their potential. These yeasts are commonly introduced into dairy products, including kefir, yogurt, kumis, cheese. However, the compatibility matrices, well factors influencing viability functionality, remains critical area study. Ongoing exploring techniques to enhance during processing, given that different strains may exhibit varying survival rates properties. Consequently, represent promising avenue augmenting health benefits products. This development prompts essential discussions regarding selecting suitable specific applications. paper comprehensively examines historical discovery, types, properties, applications It aims shed light effects while addressing challenges associated appropriate optimize enhancement Graphical

The development of the gut microbiome is crucial to human health, particularly during first three years life. Given its role in immune development, disturbances establishment process may have long term consequences. This review summarizes evidence for these claims, highlighting compositional changes this critical period life as well factors that affect development. Based on and animal data, we conclude early-life a determinant long-term impacting physiological, metabolic, processes. field faces challenges. Some challenges are technical, such lack standardized stool collection protocols, inconsistent DNA extraction methods, outdated sequencing technologies. Other methodological: small sample sizes, longitudinal studies, poor control confounding variables. To address limitations, advocate more robust research methodologies better understand microbiome's health disease. Improved methods will lead reliable studies deeper understanding impact outcomes.

Gut microbiome maturation in infants born prematurely is uniquely influenced by the physiological, clinical, and environmental factors surrounding preterm birth early life, leading to altered patterns of microbial succession relative term during first months life. These differences composition are implicated acute clinical conditions that disproportionately affect infants, including necrotizing enterocolitis (NEC) late-onset sepsis (LOS). Probiotic supplementation initiated life an effective prophylactic measure for preventing NEC, LOS, other concerns relevant infants. Gestational age (GA) and associated level of gastrointestinal tract maturation are major factors driving the initial gut microbiota composition in preterm infants. Besides, compared to term infants, premature infants often receive antibiotics treat infections probiotics restore optimal microbiota. How GA, antibiotics, modulate microbiota's core characteristics, resistome mobilome, remains nascent. We analysed metagenomic data from a longitudinal observational study six Norwegian neonatal intensive care units describe bacterial varying GA receiving different treatments. Short chain fatty acids (SCFAs), the principle end-products produced by anaerobic gut microbial fermentation of complex carbohydrates (CHO) in colon perform beneficial roles metabolic health. Butyrate, acetate and propionate are main SCFA metabolites, which maintain homeostasis host immune responses, enhance barrier integrity reduce inflammation via a range epigenetic modifications DNA/histone methylation underlying these effects. The infant microbiota composition is characterized higher abundances SCFA-producing bacteria. A large number vitro/vivo studies have demonstrated therapeutic implications bacteria inflammatory diseases, such as obesity asthma, but application its metabolite SCFAs to necrotizing enterocolitis (NEC), an acute necrosis distal small intestine/colon affecting premature newborns, scarce. Indeed, health effects attributed neonatal NEC still be understood. Thus, this literature review aims summarize available evidence on potential using PubMed/MEDLINE database.

Approximately 10% of individuals diagnosed with Clostridium difficile infection (CDI) show the resistance to fecal microbiota transplantation (FMT), underlying mechanisms remaining elusive. Deciphering intricate microbiome profile within this particular subset FMT-refractory patients via clinical FMT investigations assumes paramount importance, as it holds key designing targeted therapeutic interventions tailored for CDI, particularly recurrent CDI (rCDI). A cohort twenty-three afflicted rCDI, exhibiting congruent baselines, was meticulously selected FMT. Rigorous screening thousands healthy identified ten donors who met stringent health standards, while a total 171 stool samples were collected serve controls. To assess influence dynamics on efficacy, from four over continuous period twenty-five weeks. After treatment, seven exhibited an inadequate response These non-remission displayed significant reduction in α-diversity indexes. Meanwhile, prior FMT, abundance butyrate-producing Firmicutes bacteria, including Christensenellaceae_R_7_group, Ruminococcaceae_unclassified, Coprococcus_2, Fusicatenibacter, Oscillospira, and Roseburia, depleted patients. Moreover, Burkholderiales_unclassified, Oscillospira failed colonize both pre- post-treatment. Conversely, favorable higher relative Veillonella whereas its depletion commonly observed individuals. Genera interactions lower effectiveness more similar those patients, frequently these donors. Older not conducive colonization Veillonella, consistent their poor prognosis after rCDI distinct characteristics that hindered beneficial microbes. findings hold promise advancing precision therapy
