The production and weathering processes of nanoparticles (NPs) introduce charged functional groups on their surface. The neurological implications of micro- and nanoplastic exposure have recently come under scrutiny due to the environmental prevalence these synthetic materials. Parkinson's disease (PD) is a major disorder clinically characterized by intracellular Lewy-body inclusions dopaminergic neuronal death. These pathological hallmarks PD, according Braak's hypothesis, are mediated afferent propagation α synuclein (αS) via enteric nervous system, or so-called gut-brain axis. Transactive response (TAR) DNA-binding protein 43 (TDP-43) is a critical RNA/DNA-binding involved in various cellular processes, including RNA splicing, transcription regulation, and stability. Mislocalization aggregation of TDP-43 the cytoplasm are key features pathogenesis several neurodegenerative diseases, amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), Alzheimer's disease (AD). This review provides comprehensive retrospective prospective analysis research, highlighting structural insights, significant milestones, evolving understanding its physiological pathological functions. We delineate five major stages from initial discovery as hallmark neurodegeneration to recent advances liquid-liquid phase separation (LLPS) behavior interactions with processes. Furthermore, we assess therapeutic strategies targeting pathology, categorizing approaches into direct indirect interventions, alongside modulating aberrant LLPS. propose that future research will focus on three areas: polymorphisms for disease-specific therapeutics, exploring dual temporal-spatial modulation TDP-43, advancing nano-therapy. TDP-43 is an abundant and ubiquitously expressed nuclear protein that becomes dysfunctional in a spectrum of neurodegenerative diseases. Evidence suggests that exposure to organophosphate pesticides increases the risk of neurodegenerative diseases, but mechanisms remain unclear. This study investigated effects malathion on Alzheimer's disease (AD)-like symptoms at environmentally relevant concentrations using wild-type (WT) and APP/PS1 transgenic mouse models. Results showed induced AD-like cognitive impairment, amyloid-β (Aβ) accumulation, neuroinflammation in WT mice, with worsened mice. Mechanistic studies revealed gut microbiota dysbiosis (reduced Lactobacillus Akkermansia, increased Dubosiella), causing barrier impairment tryptophan metabolism disruptions. resulted a significant increase indole derivatives activation colonic aryl hydrocarbon receptor (AhR), promoting kynurenine (KYN) pathway while inhibiting serotonin (5-HT) pathway. Increased neurotoxic KYN metabolites (3-hydroxykynurenine quinolinic acid) triggered systemic inflammation, upregulating hippocampal IL-6 IL-1β mRNA levels thereby neuroinflammation. Gut disruptions caused neurotransmitter imbalances, reducing 5-HT its derivatives. These promoted AD progression both highlights crucial role microbiota-gut-brain axis by exposure, providing insights into risks posed pesticides.
