ABSTRACT Macroscale functional gradient techniques provide a continuous coordinate system that extends from unimodal regions to transmodal higher‐order networks. However, the alterations of these gradients in AD and their correlations with cognitive terms gene expression profiles remain be established. Abstract Functional interactions between brain regions can be viewed as a network, enabling neuroscientists to investigate function through network science. Here, we systematically evaluate 768 data-processing pipelines for reconstruction from resting-state functional MRI, evaluating the effect of parcellation, connectivity definition, and global signal regression. Our criteria seek that minimise motion confounds spurious test-retest discrepancies topology, while being sensitive both inter-subject differences experimental effects interest. Abstract Isolated REM Sleep Behavior Disorder (iRBD) is considered a prodrome of Parkinson’s disease (PD). We investigate whether the potentially disease-modifying compound acetyl- DL -leucine (ADLL; 5 g/d) has an effect on prodromal PD progression in 2 iRBD-patients. Outcome parameters are RBD-severity sum-score (RBD-SS-3), dopamine-transporter single-photon emission computerized tomography (DAT-SPECT) and metabolic “Parkinson-Disease-related-Pattern (PDRP)”-z-score 18 F-fluorodeoxyglucose positron (FDG-PET). After 3 weeks ADLL-treatment, RBD-SS-3 drops markedly both patients remains reduced for >18 months ADLL-treatment. In patient 1 (female), DAT-SPECT putaminal binding ratio (PBR) decreases years pretreatment from normal (1.88) to pathological (1.22) patient’s FDG-PET-PDRP-z-score rises 1.72 3.28 (pathological). 22 DAT-SPECT-PBR increases 1.67 stabilizes at 3.18. The lifespan growth of the functional connectome remains unknown. Here, we assemble task-free and structural magnetic resonance imaging data from 33,250 individuals aged 32 postmenstrual weeks to 80 years 132 global sites. We report critical inflection points in nonlinear curves mean variance connectome, peaking late fourth third decades life, respectively. After constructing a fine-grained, lifespan-wide suite system-level brain atlases, show distinct maturation timelines for segregation within different systems. Lifespan regional connectivity is organized along primary-to-association cortical axis. The clinical manifestation of Parkinson's disease (PD) appears when neurodegeneration is already advanced, compromising the efficacy disease-modifying treatment approaches. In present study, by using a mouse model obtained subchronic with neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and clearance inhibitor probenecid (MPTPp), we identified prodromal markers combining in vivo positron emission tomography (PET) imaging ex immunohistochemistry. Longitudinal PET dopamine transporter (DAT) [18F]-N-(3-fluoropropyl)-2β-carboxymethoxy-3β-(4-iodophenyl) nortropane ([18F]-FP-CIT), brain glucose metabolism 2-deoxy-2-[18F]-fluoroglucose ([18F]-FDG) were performed before MPTPp after 1, 3, 10 administrations, order assess relation between neuron integrity connectivity. results show that [18F]-FP-CIT dorsal striatum was not modified first administration MPTPp, tended decrease 3 significantly decreased administrations. Abstract BACKGROUND Alzheimer's disease (AD) is a neurodegenerative condition characterized by cognitive decline. To date, the specific dysfunction in brain's hierarchical structure AD remains unclear. METHODS We introduced structural decoupling index (SDI), based on multi‐site data set comprising functional and diffusion‐weighted magnetic resonance imaging from 793 subjects, to assess their brain hierarchy. RESULTS Compared normal controls (NCs), individuals with exhibited increased SDI within posterior superior temporal sulcus, insular gyrus, precuneus, hippocampus, amygdala, postcentral cingulate gyrus; meanwhile, patients demonstrated decreased frontal lobe.
