Atherosclerosis is an inflammatory disease of the large arteries that major cause cardiovascular (CVD) and stroke. Here, we review current understanding molecular, cellular, genetic, environmental contributions to atherosclerosis, from both individual pathway systems perspectives. We place emphasis on recent developments, some which have yielded unexpected biology, including previously unknown heterogeneity smooth muscle cells in atherosclerotic lesions, roles for senescence clonal hematopoiesis, links gut microbiome.

Abstract Microbiome studies have become routine in biomedical, agricultural and environmental sciences with diverse aims, including diversity profiling, functional characterization, translational applications. The resulting complex, often multi-omics datasets demand powerful, yet user-friendly bioinformatics tools to reveal key patterns, important biomarkers, potential activities. Here we introduce MicrobiomeAnalyst 2.0 support comprehensive statistics, visualization, interpretation, integrative analysis of data outputs commonly generated from microbiome studies. Compared the previous version, features three new modules: (i) a Raw Data Processing module for amplicon processing taxonomy annotation that connects directly Marker Profiling downstream statistical analysis; (ii) Metabolomics help dissect associations between community compositions metabolic activities through joint paired metabolomics datasets; (iii) Statistical Meta-Analysis identify consistent signatures by integrating across multiple Other improvements include added multi-factor differential interactive visualizations popular graphical outputs, updated methods prediction correlation analysis, expanded taxon set libraries based on latest literature. Millions of years co-evolution between animals and their associated microbial communities have shaped diversified the nature relationship. Studies continue to reveal new layers complexity in host–microbe interactions, fate which depends on a variety different factors, ranging from neutral processes environmental factors local dynamics. Research is increasingly integrating ecosystem-based approaches, metagenomics mathematical modelling disentangle individual contribution ecological microbiome evolution. Among all non-communicable diseases, Cardiovascular Diseases (CVDs) stand as the leading global cause of mortality. Within this spectrum, Myocardial Infarction (MI) strikingly accounts for over 15 % deaths. The intricate web risk factors MI, comprising family history, tobacco use, oral health, hypertension, nutritional pattern, and microbial infections, is firmly influenced by human gut microbiota, their diversity, richness, dysbiosis, along with respective metabolites. Host genetic factors, especially allelic variations in signaling inflammatory markers, greatly affect progression or severity disease. Host genetics can shape microbiome composition, but to what extent it does, remains unclear. Like any other complex trait, this important question be addressed by estimating the heritability (h2) of microbiome-the proportion variance in abundance each taxon that is attributable host genetic variation. However, unlike most traits, typically based on relative data, where taxon-specific abundances are expressed as total microbial a sample.We derived an analytical approximation for one obtains when using such relative, and not absolute, abundances, underlying quantitative model absolute abundances.
