Recent advances in optical clearing and light-sheet microscopy have provided unprecedented access to structural molecular information from intact tissues. However, current microscopes imposed constraints on the size, shape, number of specimens, compatibility with various protocols. Here we present a multi-immersion open-top microscope that enables simple mounting multiple specimens processed variety protocols, which will facilitate wide adoption by preclinical researchers clinical laboratories. We present an oblique plane microscope (OPM) that uses a bespoke glass-tipped tertiary objective to improve the resolution, field of view, and usability over previous variants. Owing its high numerical aperture optics, this achieves lateral axial resolutions are comparable square illumination mode lattice light-sheet microscopy, but in user friendly versatile format. Given performance, we demonstrate high-resolution imaging clathrin-mediated endocytosis, vimentin, endoplasmic reticulum, membrane dynamics, Natural Killer-mediated cytotoxicity. Therefore, for various reasons, we use methods save as much possible on this data, regardless its different forms, texts, pictures, videos, etc. In paper, mainly rely storing basic image which should protected another after changing formal composites using DWT wavelet transform. Abstract Motivation Stitching microscope images into a mosaic is an essential step in the analysis and visualization of large biological specimens, particularly human animal tissues. Recent approaches to highly multiplexed imaging generate high-plex data from sequential rounds lower-plex imaging. These methods promise yield precise molecular single-cell information on cellular neighborhoods tissue architecture. However, attaining with accuracy requires robust image stitching registration capabilities that are not met by existing methods. Results We describe development testing ASHLAR, Python tool for coordinated 103 or more individual accurate whole-slide mosaics. ASHLAR reads formats most commercial microscopes slide scanners, we show it performs better than open-source software. outputs standard OME-TIFF ready other tools recently developed pipelines.
