Surface Plasmon Resonance (SPR)-based biodetection systems have emerged as powerful tools for real-time, label-free biomolecular interaction analysis, revolutionizing fields such diagnostics, drug discovery, and environmental monitoring. This review highlights the foundational principles of SPR, focusing on interplay evanescent waves surface plasmons that underpin its high sensitivity specificity. Recent advancements in SPR technology, including enhancements sensor chip materials, integration with nanostructures, coupling complementary detection techniques, are discussed to showcase their role improving analytical performance. The paper also explores diverse applications systems, ranging from pathogen cancer biomarker identification food safety monitoring toxin analysis. By providing a comprehensive overview technological progress emerging trends, this underscores transformative potential SPR-based addressing critical scientific societal challenges. Optical biosensors exhibit immense potential, offering extraordinary possibilities for biosensing due to their high sensitivity, reusability, and ultrafast sensing capabilities. This review provides a concise overview of optical biosensors, encompassing various platforms, operational mechanisms, underlying physics, it summarizes recent advancements in the field. Special attention is given plasmonic metasurface-based emphasizing significant performance bioassays and, thus, increasing attraction research, positioning them as excellent candidates lab-on-chip point-of-care devices. For we emphasize surface plasmon resonance (SPR) its subcategories, along with localized (LSPR) devices enhance Raman spectroscopy (SERS), highlighting ability perform diverse bioassays. Additionally, discuss recently emerged biosensors. Toward conclusion this review, address current challenges, opportunities, prospects biosensing. Considering advantages presented by foreseeable that they will become robust widespread platform early disease diagnostics.
