En cas d’urgence radiologique ou nucléaire résultant d’un accident acte de malveillance, la prise en charge médicale requiert l’identification et le diagnostic des individus exposés à fortes doses rayonnements ionisants plus rapidement possible. Bien qu’un triage préliminaire puisse être effectué directement sur terrain, une analyse complémentaire laboratoire est nécessaire pour affiner diagnostic. Les techniques actuellement utilisées limitent rapide simultanée nombreux patients. Afin pallier ces contraintes, l’utilisation nouveaux biomarqueurs prédire risque gravité lésions radio-induites l’étude. Dans cette revue, nous abordons potentiel micro-ARN comme pronostic leur pertinence utilisation radioprotection chez l’homme.

A radiological accident may result in the development of a local skin radiation injury (LRI) which evolve, depending on dose, from dry desquamation to deep ulceration and necrosis through unpredictable inflammatory waves. Therefore, early diagnosis victims LRI is crucial for improving medical care efficiency. This preclinical study aims identify circulating metabolites as biomarkers associated with using C57BL/6J mouse model hind limb irradiation. More precisely, two independent mice cohorts were used conduct broad-spectrum profiling followed by suspect screening analysis performed plasma mass spectrometry. An integrative was conducted multi-block sparse partial least square discriminant (sPLS-DA) establish multi-scale correlations between specific levels biological, physiological (injury severity), functional parameters (skin perfusion). The identified biomarker signature consists 6-metabolite panel including putrescine, uracil, 2,3-dihydroxybenzoate, 3-hydroxybenzoate, L-alanine pyroglutamate, that can discriminate according dose severity. Our results demonstrate relevant molecular support use suitable prognosis diagnosis.

Abstract Kidney transplantation is considered the benchmark treatment for end-stage kidney disease patients.
