The significantly rising incidence of oral cancer worldwide urgently requires the identification novel, effective molecular targets to inhibit progression malignancy. DNA topoisomerase I (Topo I) is a well-established target for treatment, and many studies have shown that different cell genes could be targeted more selectively with one type Topo inhibitor. In this report, new scaffold pyridothieno[3,2-c]isoquinoline 11,11-dioxide was designed via combination key fragment or bioisoster inhibitor azaindenoisoquinolines G-quadruplex binder quindoline. Thirty-two derivatives were synthesized, among which compounds 7be, potent inhibition, exhibited antiproliferative activity against Cal27, lines highly expressing protein. Further indicated 7be also activation PI3K/AKT/NF-κB pathway downregulate level c-MYC, repress colony formation migration Cal27 cells trigger apoptosis autophagy. Molecular docking interact complex mode similar indenoisoquinolines. results xenograft model confirmed promising anticancer efficacy in vivo, tumor growth inhibition (TGI) 64.7% at 20 mg/kg.

Aim: The present study explored the bone regeneration potential of chitosan-thiocolchicoside-lauric (CTL) nanogel using MG-63 cell lines. cytocompatibility CTL was also studied osteoblast-like cells (MG-63 lines) and zebrafish embryos. Methods: effect on metabolic wound-healing activity investigated in study. alkaline phosphatase (ALP) sialoprotein (BSP) nanogel-treated were assessed ELISA. RUNX2, ALP, BSP, COL1A1 gene expression after treatment with nanogel. Hatching rates viability embryos treated different concentrations studied. Any developmental toxicity investigated. Results: There no significant reduction proliferation when 5–20 μL/mL MTT assay (p < 0.05). No prominent morphological changes or nuclear abnormalities found various compared to control group. Invitro scratch wound healing showed excellent migration and, hence, ability ELISA ALP BSP cells. significantly increased group hatching as decreased, highest rate observed at a 5 µL concentration. Developmental toxicity, such spinal cord bent, tail yolk sac oedema, not Conclusions: can be used treating defects helping healing. suggests that had concentration-dependent embryos’ only very high concentrations.

Oral squamous cell carcinoma (OSCC) is a disease of significant concern with higher mortality rates. Conventional treatment approaches have several drawbacks, leading to the opening new research avenues in field nanoparticle-based cancer therapeutics. The study aimed at synthesis gold nanoparticles (Pm-AuNPs) from aqueous bark extract Pterocarpus marsupium, followed by its characterization and vitro anticancer evaluation against OSCC. synthesized Pm-AuNPs were characterized using UV–visible spectroscopy, particle size analyser, zeta potential, FTIR SEM techniques. potential was evaluated OSCC lines (SCC29b, SSC154 OECM-1) through assays. IC50 value found be 25 ± 1.2, 45 1.5 75 2.1 µg/mL for three lines, elucidating cytotoxic effects mechanism action. Intracellular ROS SOX detection, mitochondrial transmembrane analysis apoptosis detection used confirm activity Acute toxicity studies on Wistar rats confirmed non-toxic nature dose concentration up 2000 mg/kg body weight. findings underscore as promising candidates future therapeutics, providing insights into their action therapeutic efficacy
