Abstract Many experts have extensively studied the potential of exercise as a treatment option for psychiatric conditions, including depression and autism spectrum disorder (ASD). Despite their core symptoms, these conditions exhibits comparable component traits, an anxiety. In this study, we explored effect on behavioral abnormalities in focusing its intensity emotional resilience. Shank3B knockout (KO SED ) mice displaying self-injurious repetitive behavior C57BL/6J mice, susceptible to stress ASD model, respectively, were subjected moderate-intensity (ME) 2 weeks. ME mitigated symptoms (excessive grooming traits despair) but did not exert significant anxiolytic effect. Notably, has emerged critical determinant efficacy, evidenced by lower ventilation threshold mediated low-intensity exercise. The findings substantiate notion that is promising disease-modifying treatment, matters Graphical

Abstract Closed head injury is a prevalent form of traumatic brain with poorly understood effects on cortical neural circuits. Given the emotional and behavioral impairments linked to closed injury, it vital uncover functional deficits their driving mechanisms. In this study, we employed robust viral tracing technique identify alteration pathway connecting medial prefrontal cortex basolateral amygdala, observed disruptions in neuronal projections between amygdala following injury. Remarkably, our results highlight that ZL006, an inhibitor targeting PSD-95/nNOS interaction, stands out for its ability selectively reverse these aberrations. Specifically, ZL006 effectively mitigates from induced by Furthermore, using chemogenetic approaches, elucidate activating circuit produces anxiolytic effects, aligning therapeutic potential ZL006. Additionally, administration astrocyte activation, leading restoration glutamatergic neuron activity. Moreover, context attenuating anxiety-like behaviors through treatment, observe reduction injury–induced engulfment, which may correlate decrease dendritic spine density neurons.
