Metabolomics plays a pivotal role in systems biology, and NMR is central tool with high precision exceptional resolution of chemical information. Most metabolomic studies are based on 1H 1D spectroscopy, severely limited by peak overlap. 13C benefits from larger signal dispersion but barely used metabolomics due to ca. 6000-fold lower sensitivity. We introduce new approach, hyperpolarized at natural abundance, that circumvents this limitation. A untargeted NMR-based workflow dissolution dynamic nuclear polarization (d-DNP) for the first time enabled abundance metabolomics. The combination of ion-mobility (IM) separation with mass spectrometry (MS) has impacted global measurement efforts in areas ranging from food analysis to drug discovery. Reasons for the broad adoption IM-MS include its significantly increased peak capacity, duty-cycle, and ability reconstruct fragmentation data parallel, all which greatly enable analyses complex mixtures. More fundamentally, however, measurements ion-gas molecule collision cross sections (CCSs) are used support compound identification quantitation as well study structures large biomolecules. As first commercialized form IM-MS, Traveling Wave Ion Mobility (TWIM) devices operated at low pressures (∼3 mbar) voltages, relatively short (∼25 cm), separate ions on a timescale tens milliseconds. These qualities make TWIM ideally suited hybridization MS. Owing complicated motion devices, IM transit times must be calibrated CCS measurements.
