Surface-enhanced Raman scattering (SERS) and tip-enhanced (TERS) have opened a variety of exciting research fields. However, although vast number applications been proposed since the two techniques were first reported, none has applied to real practical use. This calls for an update in recent fundamental application studies SERS TERS. Thus, goals scope this review are report new directions perspectives TERS, mainly from viewpoint combining their mechanism studies. Regarding progress discusses four main topics: (1) nanometer subnanometer plasmonic hotspots SERS; (2) Ångström resolved TERS; (3) chemical mechanisms, i.e., charge-transfer semiconductor-enhanced scattering; (4) creation strong bridge between applications.

Phototherapy of deep tumors still suffers from many obstacles, such as limited near-infrared (NIR) tissue penetration depth and low accumulation efficiency within the target sites. Herein, stimuli-sensitive tumor-targeted photodynamic nanoparticles (STPNs) with persistent luminescence for treatment are reported. Purpurin 18 (Pu18), a porphyrin derivative, is utilized photosensitizer to produce in STPNs, while lanthanide-doped upconversion (UCNPs) exhibit bioimaging properties possess high photostability that can enhance efficacy. STPNs initially stimulated by NIR irradiation before intravenous administration accumulate at tumor site enter cells through HER2 receptor. Due Pu18 afterglow properties, continuously generate ROS inhibit NFκB nuclear translocation, leading cell apoptosis. Cells and their derived extracellular vesicles (EVs) or exosomes contain unique molecular signatures that could be used as biomarkers for the detection of severe diseases such cancer, well monitoring treatment response. Revealing these requires developing non-invasive ultrasensitive tools to enable single molecule/cell-level using a small volume sample with low signal-to-noise ratio background multiplex capability. Surface-enhanced Raman scattering (SERS) can address current limitations in studying cells EVs through two main mechanisms: plasmon-enhanced electric field (the so-called electromagnetic mechanism (EM)), chemical (CM). In this review, we first highlight SERS mechanisms then discuss nanomaterials have been develop biosensors based on each aforementioned combination order take advantage synergic effect between enhancement enhancement. Metal nitride nanostructures have been predicted to exhibit plasmonic responses in the UV IR region, which can enable their potential use as low-cost, chemically, and thermally stable materials several applications.
