Enzymes and cell factories play essential roles in industrial biotechnology for the production of chemicals fuels. The properties natural enzymes cells often cannot meet requirements different processes terms cost-effectiveness high durability. Droplet-based microfluidics has been widely applied in enzyme directed evolution (DE), either cell or cell-free system, due to its low cost and high throughput. As the isolation principles are based on labeled label-free characteristics droplets, sorting method contributes mostly efficiency of whole system. Fluorescence-activated droplet (FADS) is but faces challenges target scope. Label-free methods show potential greatly broaden microfluidic application range. Here, we review developments through a comprehensive literature survey, including detections [FADS absorbance-activated (AADS)] [electrochemical-based (ECDS), mass-activated (MADS), Raman-activated (RADS), nuclear magnetic resonance-based (NMR-DS)]. We highlight recent cases last 5 years which novel enzymes highly efficient variants generated by DE. In addition, advantages different briefly discussed provide an outlook for future applications

Accurate and efficient droplet microfluidic strategy for controlling the morphology of energetic microspheres DOI

An effective method is proposed for preparing energetic microspheres by fluid-focusing microchannels. This combines the advantages of low reagent consumption, instrument miniaturization, accurate control, and efficient heat mass transfer. The effects two-phase liquid flow rate ratio water to oil on droplet formation was fully studied. To verify applicability method, nitroamine explosives HMX (cyclotetramethylene tetranitramine) employed with fluorine rubber (F2602) as binder prepare HMX/F2602 microsphere, which were compared samples prepared traditional water-suspension method. SEM data showed that morphology, particle size uniformity, monodispersity microfluidic are better than those DSC suspension apparent activation energy (Ea) values increased 25.47 kJ/mol coated sample. Meanwhile, raw material HMX, H50, fluidity bulk density significantly improved (HMX/F2602 microsphere HMX: H50 = 86.9 25.2 cm, repose angle 33.1 51.3°, 0.714 0.604 g cm−3). study demonstrates feasibility an efficient, strategy explosive microspheres.
