Droplet-based
microfluidics
has
been
widely
applied
in
enzyme
directed
evolution
(DE),
either
cell
or
cell-free
system,
due
to
its
low
cost
and
high
throughput.
As
the
isolation
principles
are
based
on
labeled
label-free
characteristics
droplets,
sorting
method
contributes
mostly
efficiency
of
whole
system.
Fluorescence-activated
droplet
(FADS)
is
but
faces
challenges
target
scope.
Label-free
methods
show
potential
greatly
broaden
microfluidic
application
range.
Here,
we
review
developments
through
a
comprehensive
literature
survey,
including
detections
[FADS
absorbance-activated
(AADS)]
[electrochemical-based
(ECDS),
mass-activated
(MADS),
Raman-activated
(RADS),
nuclear
magnetic
resonance-based
(NMR-DS)].
We
highlight
recent
cases
last
5
years
which
novel
enzymes
highly
efficient
variants
generated
by
DE.
In
addition,
advantages
different
briefly
discussed
provide
an
outlook
for
future
applications

microspheres
