Abstract Cancer is one of the most intractable diseases in world because its high recurrence rate, metastasis rate and lethality rate. Traditional chemotherapy, radiotherapy surgery have unsatisfactory therapeutic effects cause many severe side at same time. Hydrogel a new type biomaterial with advantages good biocompatibility easy degradation, which can be used as carrier functional nanomaterials for tumor therapy. Herein, we represent progress hydrogels different skeletons their application treatment. The are listed polyethylene glycol‐based hydrogels, chitosan‐based peptide‐based hyaluronic acid‐based steroid‐based other by skeletons, properties, modifications toxicities were introduced. Some representative applications combined nanomaterial photodynamic therapy, photothermal sonodynamic chemodynamic therapy synergistic highlighted.

At present, an oral tumor is usually treated by surgery combined with preoperative or postoperative radiotherapies and chemotherapies. However, traditional chemotherapies frequently result in substantial toxic side effects, including bone marrow suppression, malfunction of the liver kidneys, neurotoxicity. As a new local drug delivery system, smart system based on hydrogel can control release time space, effectively alleviate avoid these problems. Cancer seriously threatens human health. Surgery, radiotherapy and chemotherapy are the three pillars of traditional cancer treatment, with targeted therapy immunotherapy emerging over recent decades. Standard drug regimens mostly executed via intravenous injection (IV), especially for agents. However, these treatments pose severe risks, including off-target toxic side effects, low accumulation penetration at tumor site, repeated administration, etc., leading to inadequate treatment failure meet patients’ needs. Arising from challenges, a local regional anticancer strategy has been proposed enhance therapeutic efficacy concomitantly reduce systemic toxicity. With advances in biomaterials our understanding microenvironment, situ stimulus-responsive hydrogels, also called smart have extensively investigated due their injectability, compatibility responsiveness various stimuli (pH, enzyme, heat, light, magnetic fields, electric fields etc.). The DNA damage repair of tumor cells limits the effect photothermal therapy (PTT), and high temperatures induced by PTT can adjacent normal tissues. To overcome these limitations, we developed a novel composite hydrogel (OLA-Au-Gel) based on chitosan (CS) β-glycerophosphate (β-GP), which encapsulated olaparib-liposomes (OLA-lips) CS-capped gold nanoparticles (CS-AuNPs). OLA-Au-Gel achieved combination mild (mPTT) CS-AuNPs inhibition OLA. showed good biocompatibility, injectability, response. Under near-infrared laser irradiation, inhibited proliferation cells, generation reactive oxygen species in vitro, effectively growth breast tumors vivo. shows promising application prospect for inhibiting development improving antitumor effect. Collectively, propose strategy enhanced mPTT inhibition.
