The concept of molecular design, integrating diagnostic and therapeutic functions, aligns with the general trend modern medical advancement. Tumor-associated mast cells (TAMCs) have been recently revealed to play a multifaceted role in the tumor microenvironment. Noninvasive optical imaging of TAMCs is thus highly desired gain insights into their functions cancer immunotherapy. However, due lack single enzyme that specific cells, common probe design approach based on single-enzyme activation not applicable. Herein, we reported bienzyme-locked molecular (THCMC) photoinduced electron transfer-intramolecular charge-transfer hybrid strategy for vivo TAMCs. The mechanism ensures THCMC exclusively turns near-infrared (NIR) fluorescence only presence both tryptase and chymase specifically coexpressed by cells. Thus, effectively distinguishes from other leukocytes, including T neutrophils, macrophages, capability lacking single-locked probes. Such high specificity allows noninvasive tracking fluctuation living mice during results reveal decreased intratumoral signal after combination immunotherapy correlates well with reduced population TAMCs, accurately predicting inhibition growth. this study presents first NIR fluorescent but also proposes generic cell imaging.

Intracranial optical imaging of glioblastoma (GBM) is challenging due to the scarcity effective probes with blood-brain barrier (BBB) permeability and sufficient depth. Herein, we describe a rational strategy for designing crossing BBB based on an electron donor-π-acceptor system adjust lipid/water partition coefficient molecular weight probes. The amphiphilic hemicyanine dye (namely, IVTPO), which exhibits remarkable properties permeability, chosen as efficient fluorescence/photoacoustic probe in vivo real-time orthotopic GBM high resolution through intact skull. Abnormal leakage IVTPO adjacent developing tumor unambiguously observed at early stage development prior impairment integrity, assessed by commercial Evans blue (EB). Compared EB, demonstrates enhanced capability improved tumor-targeting efficacy. These results offer encouraging insights into medical diagnosis intracranial GBM.

Accurate and selective techniques for visualizing endogenous peroxynitrite (ONOO-) in cerebral ischemia-reperfusion injury (CIRI) models are essential understanding its complex pathological processes. Here, we introduced a longwave fluorescent probe TJO detecting ONOO- rapidly sensitively, with low detection limit of 91 nM. Furthermore, exhibits excellent fluorescence imaging capabilities, enabling detailed visualization ONOO⁻ CIRI mice model. This highlights potential real-time monitoring ONOO⁻-related conditions.
