A flow-through biosensor system for the determination of uric acid was developed on platform electrochemical cell manufactured by 3D printing from poly(lactic acid) and equipped with a modified screen-printed graphite electrode (SPE). Uricase immobilized to inner surface replaceable reactor chamber. Its working volume reduced 10 μL against previously reported similar cell. SPE independently enzyme carbon black, pillar[5]arene, poly(amidoamine) dendrimers based p-tert-butylthiacalix[4]arene (PAMAM-calix-dendrimers) electropolymerized 3,7-bis(4-aminophenylamino) phenothiazin-5-ium chloride. Introduction PAMAM-calix-dendrimers into coating led fivefold increase in redox currents electroactive polymer. It found that higher generations greater measured. Coatings consisted products electropolymerization phenothiazine implemented pillar[5]arene showing high efficiency reduction hydrogen peroxide formed enzymatic oxidation acid. The presence PAMAM-calix-dendrimer G2 increased signal related assay more than 1.5 times. successfully applied chronoamperometric mode. following optimal parameters conditions were established: pH 8.0, flow rate 0.2 mL·min−1, 5 U uricase per reactor. Under these conditions, made it possible determine nM 20 μM limit detection (LOD) 4 nM. Glucose (up 1 mM), dopamine 0.5 ascorbic 50 μM) did not affect toward tested spiked artificial urine samples, showed 101% recovery tenfold diluted samples. ease assembly low cost replacement parts make promising future application routine clinical analyses.
