The use of combination therapies that employ a variety cell death mechanisms has emerged as promising avenue research in the treatment cancer. However, optimization therapeutic synergies when integrating different modes remains significant challenge. To this end, we developed multifunctional intelligent drug-carrying nanoparticle (DFMTCH NPs) based on metal-organic framework MIL-100, loaded with doxorubicin (DOX) and disulfiram (DSF), coated Cu-tannic acid (Cu-TA) network hyaluronic (HA), for purpose combined chemotherapy/chemodynamic/photothermal anti-cancer therapy. On one hand, DFMTCH NPs exhibited range capabilities, including chemotherapy, photothermal therapy (PTT), chemodynamic (CDT), which collectively enhanced anti-tumor efficacy chemotherapeutic agents. In addition, proved sensitive photoacoustic imaging (PAI) image-guided other could produce reactive oxygen species (ROS) consume glutathione (GSH) by amplifying cellular oxidative stress, while causing intracellular mitochondrial dysfunction, inducing effective cuproptosis/ferroptosis/apoptosis to inhibit tumor growth. Collectively, work provided an innovative strategy designing nanoparticles combat colorectal cancer (CRC).
