Whey protein is widely recognized as a high-quality source, primarily derived from dairy products. This type of valued not only for its remarkable nutritional benefits but also substantial antioxidant properties and capacity to reduce inflammation. Previous studies have demonstrated that whey hydrolysate (WPH) effective in both the prevention treatment liver injury caused by acetaminophen (APAP) overdose. Pyroptosis form programmed cell death significantly contributes damage induced APAP, it associated with increased levels oxidative stress However, specific mechanisms which WPH influences pyroptosis remain fully understood. The present study observed significant alleviation APAP-induced hepatocyte WPH, alongside reduction release pro-inflammatory factors, specifically interleukin-1β (IL-1β) interleukin-18 (IL-18). Moreover, expression pyroptosis-related proteins, including NOD-like receptor family pyrin domain containing 3 (NLRP3), N-terminal gasdermin D (GSDMD-N), cleaved caspase-1, were reduced following WPH. In subsequent phase our research, we examined reactive oxygen species (ROS) cathepsin B (CTSB) hepatocytes. Increased ROS resulted heightened permeability lysosomal membranes, led CTSB into cytosol. liberated subsequently activated NLRP3 inflammasome, thereby promoting pyroptosis. Our results revealed accumulation hepatocytes inhibited CTSB. mechanistic studies, found upregulated Parkin, PTEN-induced putative kinase 1 (PINK1), microtubule-associated light chain β (LC3), while downregulating P62 activate mitophagy. Apigenin (API) is a flavonoid widely distributed in vegetables and fruits that exhibits numerous biological functions. Lipopolysaccharide (LPS), key component of the outer membrane Gram-negative bacteria, can cause kidney injury when released into bloodstream. Necroptosis form programmed cell death characterized by rupture membranes. Excessive occurrence necroptosis lead to substantial damage cells tissues. In study, we discovered API could mitigate LPS-induced mice alleviate Normal Rat Kidney-52E (NRK-52E) targeting mitochondrial reactive oxygen species (mtROS)-RIPK3-MLKL pathway. Further mechanistic studies revealed potentially activate endosomal sorting complexes required for transport-III (ESCRT-III), activated ESCRT-III repair caused necroptosis. Simultaneously, promote mitophagy, which facilitates timely removal damaged mitochondria reduces intracellular mtROS levels. conclusion, our results suggested alleviates renal activating ESCRT-III-dependent mitophagy.
