Abstract Regulation of human papillomavirus (HPV) gene expression is tightly linked to differentiation the keratinocytes virus infects. HPV late confined cells in upper layers epithelium where capsid proteins are synthesized. As these highly immunogenic, and an immune‐privileged site, this spatial restriction aids immune evasion. Many decades work have contributed current understanding how occurs at a molecular level. This review will examine what known about HPV‐infected lesions dissect intricacies regulation. Future directions for novel antiviral approaches be highlighted.

Significance Human papillomaviruses (HPV) uncouple proliferation from differentiation to enable virus replication in epithelial cells. HPV E7 proteins are well established promote by binding and inactivating retinoblastoma family other cell cycle inhibitors. However, mechanisms which high-risk oncoproteins inhibit have not been defined. This paper identifies a mechanism keratinocyte differentiation. The inhibition of requires degradation the cellular protein PTPN14 E7, this is related ability immortalize keratinocytes cause cancer.

Objective The monoclonal antibodies anti-programmed death protein-1 (anti–PD-1) nivolumab and pembrolizumab are the first immune checkpoint inhibitors (ICIs) approved for treatment of recurrent/metastatic head neck carcinoma R/M HNSCC in line platinum refractory disease. This network meta-analysis aims to investigate efficacy anti–PD-1- vs anti–PD-L1-based therapy cancer patients through a systematic review literature provide support evidence-based decisions. In particular, effectiveness ICIs is analyzed according different mechanisms action check-points inhibitory drugs subgroups patients. Methods We did (NMA) randomized controlled trials (RCTs) PubMed, ClinicalTrials.gov, Embase, Medline, Cochrane Central Register Controlled Trials, Web Science. Our search identified total five trials: Keynote 040, 048, Eagle, Condor, Checkmate 141. These included 3001 Treatment was sub-categorized into PD-L1–based, PD-1–based, standard chemotherapy. Treatments were indirectly compared with therapy. Results demonstrated no significant differences OS between except metastatic which anti–PD-1-based associated significantly less risk death. Furthermore, appeared be effective smoker human papilloma–negative (HPV) Conversely, seems better efficient female patients, locally recurrent setting HPV positive Conclusion NMA study that aimed compare results our could help define profile patient responder or resistant specific classes can used guide/design future studies novel scenario precision immune-oncology.

Persistent infection with some types of mucosal human papillomavirus (HPV) is the etiological factor for development cervical cancer and its precursor lesions. Besides, several cofactors are known to play a role in disease onset progression either by favoring or preventing HPV persistence. The microbiome healthy female genital tract characterized presence 1 few varieties lactobacilli. However, high-throughput studies addressing bacterial diversity abundance have shown that factors, including hormonal levels, hygiene habits, sexually transmitted diseases may disrupt natural balance, outgrowth groups bacteria, which turn favor pathological states. Recently, vaginal has emerged as new variable could greatly influence history infections their clinical impact. In this context, changes been detected women infected HPV-associated lesions cancer. specific bacteria development/progression prevention/regression pathologies not well understood. review we summarize current knowledge concerning disease. Cervical cancer (CC) remains a public health issue worldwide despite preventive measures. Surgical treatment in the early-stage CC has evolved during last decades. Our aim was to review advances literature and summarize ongoing studies on this topic. To end, we conducted through PubMed focusing English-language articles surgical management of CC. The emergent topics considered here are FIGO 2018 staging system update, conservative with less radical procedures for selected patients, lymph node staging, fertility preservation, preferred approach, tumors up 2 cm, prognosis. In terms updating FIGO, highlight inclusion status possibility imaging. Regarding emphasize LACC trial impact favor open surgery; however, discuss controversial application < cm. summary, all show tendency provide patients tailored that avoids morbidity while maintaining oncologic safety, which is already possible high-income countries. We believe efforts should focus making reality low-income countries as well.

Abstract Human papillomavirus (HPV) is the most prevalent sexually transmitted virus globally. Persistent high‐risk HPV infection can result in cervical precancerous lesions and cancer, with 70% of cancer cases associated types HPV16 18. imposes a significant financial psychological burden. Therefore, studying methods to eradicate halt progression remains crucial. This review comprehensively explores mechanisms underlying HPV‐related lesions, including viral life cycle, immune factors, epithelial cell malignant transformation, host environmental contributing factors. Additionally, we provide comprehensive overview treatment for cancer. Our focus on immunotherapy, encompassing therapeutic vaccines, checkpoint inhibitors, advanced adoptive T therapy. Furthermore, summarize commonly employed drugs other nonsurgical treatments currently utilized clinical practice managing lesions. Gene editing technology undergoing research and, although not yet officially numerous preclinical studies have substantiated its efficacy. it holds promise as precise strategy

Human papillomavirus (HPV) is a group of alpha papillomaviruses that cause various illnesses, including cancer. There are more than 160 types HPV, with many being "high-risk" have been clinically linked to cervical and other "Low-risk" HPV less severe conditions, such as genital warts. Over the past few decades, numerous studies shed light on how induces carcinogenesis. The genome circular double-stranded DNA molecule approximately 8 kilobases in size. Replication this strictly regulated requires two virus-encoded proteins, E1 E2. helicase necessary for replisome assembly replication genome. On hand, E2 responsible initiating regulating transcription HPV-encoded genes, most importantly E6 E7 oncogenes. This article explores genetic characteristics high-risk types, roles proteins replication, regulation oncogenes, development oncogenesis.
