Background/Objectives: The use of antibiotics in livestock contributes to antimicrobial resistance, highlighting the need for alternative solutions. Abstract Background Antimicrobial resistance (AMR) is nowadays a major emerging challenge for public health worldwide. The increasing reliance of modern agriculture on honey bee (Apis mellifera) pollination has driven efforts to preserve and enhance populations. cryopreservation drone semen presents a promising solution for preserving genetic diversity supporting breeding programs without live animal transport risks. This study aimed evaluate one-step dilution antibiotic-free slow-freezing protocol under field conditions with in vitro vivo parameters. Semen viability was tested by two different mixes dyes, both techniques gave similar results, showing post-thaw drop 37%. Virgin queens were inseminated either fresh or frozen-thawed semen. Survival rates until egg-laying onset female brood production groups; however, colonies more likely go into wintering. ABSTRACT This study investigated the potential of incorporating curcumin oil nano‐emulsion (CONE) into a Tris‐based freezing extender to relieve cryodamage effects and enhance sperm cryo‐resistance in buffalo bulls. Pooled semen from five fertile bulls ( n = 40 ejaculates) was divided supplemented with 0.5% CONE (CONE0.5), 1% (CONE1), or without (CONE0, control), then extended packaged. Post‐thaw assessments included kinematic parameters, oxidative stress markers, apoptosis levels, microbial load, ultrastructural integrity, molecular docking interactions, conception rates. This study evaluated the impact of different processing techniques on microbial load and sperm quality in frozen–thawed equine semen to identify alternatives reduce preventive use antibiotics. Semen was obtained processed under rigorous hygiene measures from ten stallions, using four protocols: Simple Centrifugation with antibiotics (S+) (S−), Filtration (F−) Single-Layer Colloidal (C−) an antibiotic-free extender. Microbial culture media, viability motility were assessed. results consistent across protocols, except Columbia 5% Sheep Blood Agar where S− exhibited higher than S+ (p < 0.05). However, F− C− showed similar loads S+. No significant differences observed progressive motility, average path velocity, straight-line velocity or wobble parameters between protocols.
