The
circadian
timing
system
(CTS)
controls
various
biological
functions
in
mammals
including
xenobiotic
metabolism
and
detoxification,
immune
functions,
cell
cycle
events,
apoptosis
angiogenesis.
Although
the
importance
of
CTS
is
well
known
pharmacology
drugs,
it
less
appreciated
at
clinical
level.
Genome-wide
studies
highlighted
that
majority
drug
target
genes
are
controlled
by
CTS.
This
suggests
chronotherapeutic
approaches
should
be
taken
for
many
drugs
to
enhance
their
effectiveness.
Currently
successfully
applied
treatment
different
types
cancers.

Overnutrition

circadian
metabolic
rhythms
by
mechanisms

not
well
understood.

we
show
diet-induced
obesity
(DIO)
causes
massive
remodeling
of
enhancer
activity
in
mouse
liver,

synchronous
high-amplitude

fatty
acid
(FA)
synthesis

oxidation.
SREBP
expression

induced
DIO,
leading
to
FA

oxidation

required

FAO.

Abstract
Shift
work,
defined
as
work
occurring
outside
typical
daytime
working
hours,
is
associated
with
an
increased
risk
of
various
non‐communicable
diseases,
including
diabetes
and
cardiovascular
disease.
Disruption
the
internal
circadian
timing
system
concomitant
sleep
disturbances
thought
to
play
a
critical
role
in
development
these
health
problems.
Indeed,
controlled
laboratory
studies
have
shown
that
short‐term
misalignment
restriction
independently
impair
physiological
processes,
insulin
sensitivity,
energy
expenditure,
immune
function,
blood
pressure
cardiac
modulation
by
autonomous
nervous

Hepatocellular
carcinoma
(HCC)
remains
a
global
health
challenge
whose
incidence
is
growing
worldwide.
Previous
evidence

clock
controls

homeostasis

hepatocarcinogenesis.

Abstract
Background
Tolerability
and
antitumour
efficacy
of
chemotherapy
radiation
therapy
can
vary
largely
according
to
their
time
administration
along
the
24-h
scale,
due
moderation

mechanisms

circadian
rhythms.
Recent
clinical
data
have
highlighted
a
striking
role
dosing
for
cancer
immunotherapy,
thus
calling
critical
evaluation.
Methods

we

analyse

rhythms

ICI
therapies.

rhythm
disorders
affect
tumour
immune
microenvironment,

infusion

inhibitors
(ICIs)

enhance
progression-free

survival

ToD

Circadian
clocks
play

TimeSignature,

independent
studies

microarrays

validate
it
against
set
profiled

RNA-sequencing.

results
show
TimeSignature
more
accurate
efficient
than
competing
methods,
estimating
within
2
h
majority
samples.
Importantly,
once
trained
on
single
study,
predictor
yield
highly
differences
study
population,
patient
protocol,
or
assay
platform
without
renormalizing
retraining.

feature
unique
among
expression-based
addresses

generalizable,
